摘要
为提高微杆菌Z4发酵的几丁质酶得率,利用摇瓶研究了发酵温度、初始pH、装液量等对于发酵产酶的影响.在这些实验的基础上,运用正交试验对Z4产几丁质酶的摇瓶发酵条件进行了进一步优化.结果的极差分析和方差分析表明温度对产酶的影响最大,具有显著性.最佳摇瓶发酵工艺条件为:培养温度28℃,培养基初始pH7,接种量6%,三角瓶装液量60mL培养基/250 mL三角瓶.在该条件下发酵液酶产量达到4.13 U/mL,比优化前提高了79.6%.
The effect of the fermentation temperature,inoculum ratio and liquid volume of the flask on the chitinase fermentation of Microbacterium sp Z4was studied.The fermentation condition for optimized production of chitinase was further studied by using L9(34)orthogonal experimental method.The maximal chitinase activity up to 4.13U/mL was attained under the condition:temperature of 28degrees Celsius,initial pH 7,inoculums 6%,and 60mL medium in 250mL flask.The chitinase production increased 79.6%of the initial production.
