重组空肠弯曲菌蛋白疫苗免疫BALB/c小鼠的研究

摘要 目的探讨重组空肠弯曲菌rLTB-PEB1蛋白疫苗的免疫机制。方法经口服免疫 BALB/c 小鼠后,ELISA 法检测特异性抗体IgG 、IgA 水平;ELISPOT 法检测派伊尔结特异性抗体分泌细胞;实时荧光定量PCR 检测IFN-γ、IL-4 mRNA 表达的差异。计算疫苗保护率。结果免疫组小鼠派伊尔结特异性抗体、IFN-γ、IL-4 mRNA 水平明显增高( P <0.05)。免疫组的疫苗保护率为89.3%~93.7%,可维持30周。结论重组空肠弯曲菌rLTB-PEB1蛋白疫苗口服免疫小鼠可有效预防空肠弯曲菌的感染,其免疫机制可能为诱导了TH1/TH2型免疫应答。 Objective To study the immunological response induced by Campylobacter jejuni (CJ) recom-binant protein vaccine .Methods After the oral immunization for BALB/c mice ,the levels of IgG and IgA were detected by ELISA , the specific antibody secreting cells in Payer’s patch were detected by ELISPOT , the expression levels of IFN-γ and IL-4 were detected by RT-PCR .The vaccine protection rate was calculated .Results The levels of specific antibody , IFN-γ and IL-4 were increased obviously after immuni-zation .The vaccine protection rate was 89 .3%-93 .7%( P 〈 0 .05) .Conclusion The oral immunization with recombinant protein vaccine may induce TH1/TH2 type immune responses and can produce protective im-munity against Campylobacter jejuni .
