摘要
用聚丙烯酰胺凝胶电泳在坐骨神经损伤侧远端提取具有神经诱向生长作用的1 8k Da蛋白作为抗原 ,通过免疫 BAL B/ C小鼠 ,经杂交瘤技术与 ELISA法筛选 ,获取稳定分泌单克隆抗体的杂交瘤细胞株。免疫组织化学法和计算机图像处理系统显示 :生后1 ,3,7,1 4,2 1和 2 8日龄大鼠坐骨神经中 1 8k Da蛋白免疫反应的分布 。
The study of neuro chemotropism was an important aspect of neurosciences. Neurotrophic effect of the distal stump of transected sciatic nerve was purified from rat. Native polyacry lamide gel electrophoresis (PAGE) revealed the specific protein band. The monoclonal antibodies were developed through immunizing the BALB/C mouse with 18 kDa protein by assay of ELISA. The distribution of 18 kDa neurotrophic factor like immunoreactivity and the changes of the level of 18 kDa in the sciatic nerve of rats at postnatal day 1, day 3, week 1, week 2, week 3, and week 4 were studied with immunohistochemical ABC technique and computer aided image processing system. The results showed that the positive immunoreactivity was appeared in the nerves of an age groups.
