雌激素受体阴性/阳性MCF-7细胞增殖速度差异及对鸡血藤提取物敏感性的研究

摘要 目的明确雌激素受体(ER)阴性、阳性MCF-7细胞增殖速度差异以及不同MCF-7细胞对鸡血藤黄酮类提取物SSCE抗肿瘤作用的敏感性。方法免疫细胞化学(ICC)方法确定3种MCF-7细胞(MCF-7-a、MCF-7-b、MCF-7-luc)ER表达情况,MTT法检测3种细胞的增殖速度以及对SSCE抑制肿瘤细胞生长作用的敏感性。结果 ER(+)的MCF-7-a、ER(-)的MCF-7-b和MCF-7-luc倍增时间分别为(69.95±9.37)h、(47.17±5.27)h、(44.96±10.33)h,24 h、48 h、72 h各个时间点MCF-7-luc对SSCE的半数抑制浓度均低于另外两株细胞。结论 ER(-)的乳腺癌细胞恶性增殖速度高于ER(+)的乳腺癌细胞;荧光素酶基因标记的ER(-)MCF-7细胞对SSCE抗肿瘤作用更加敏感;提示ER的表达情况与乳腺癌的恶性增殖程度有关,基因重组可能影响了细胞膜表面蛋白的表达,从而增强了细胞对药物的敏感性。 Objective To clarify the differences of proliferation rate of positive or negative MCF-7 cells in estrogen receptor(ER) as well as sensibility of different MCF-7 cells on anti-tumor effect of spatholobus suberectus column extration ( SSCE). Methods The immunocytochemistry(ICC) was adapted to measure the expression of three kinds of MCF-7 cells (MCF-7-a, MCF-7-b and MCF-7- luc2) in ER. MTT method was applied to test the proliferation rate of three MCF-7 cells and their sensibility on anti-tumor effect of SSCE. Results The median inhibitory concentrations of MCF-7-1uc on SSCE at 24 h, 48 h and 72 h were (69. 95 ± 9. 37 ) h, (47.17 ± 5.27 ) h and (44. 96± 10. 33 ) h, all lowest compared with the rest two cells. Conclusion The malignant proliferation rate of ER(-) breast cancer cell is higher than that in the ER( + ) breast cancer cell. ER(-) MCF-7 cell labeled by luciferase gene has a more sensitive level on anti-tumor effect of SSCE, implying that ER expression is related with malignant proliferation of breast cancer. Genetic recombination may make an influence on expression of surface protein of eytomembrane, leading to an enhanced sensibility on pharmacy.
