三七三醇皂苷对局灶性脑缺血再灌注大鼠GFAP mRNA和蛋白表达的影响 被引量:2

摘要 目的:探讨局灶性脑缺血再灌注后梗死区周围MPA2、GFAP蛋白和mRNA表达的动态变化,以及三七三醇皂苷对其表达的影响。方法:采用改良的线栓法制备大脑中动脉阻塞45 min,不同再灌注时间(1、7、14、21、28 d)大鼠短暂局灶性脑缺血模型。应用PCR、Western动态观测微管相关蛋白、胶质纤维酸性蛋白mRNA和蛋白表达及三七三醇皂苷治疗对其影响。结果:微管相关蛋白mRNA和蛋白表达显示,再灌注1、14、21、28 d时三七三醇皂苷组、N.S组要比正常组、假手术组高,但各组之间差异均无统计学意义(P>0.05)。胶质纤维酸性蛋白mRNA和蛋白表达结果显示,再灌注后1、7、14、21、28 d三七三醇皂苷组比N.S组、正常组和假手术组高,差异均有统计学意义(P<0.05),两周时最明显。结论:脑缺血再灌注后胶质纤维酸性蛋白mRNA和蛋白表达增高,三七三醇皂苷治疗可能加速了神经干细胞的增殖,并可能使增值的神经干细胞更多向胶质细胞分化。 Objective To explore the dynamic changes in MPA2 GFAP mRNA and protein expression and the effect ofPTS on it after focal ischemia reperfusion around the infarct area. Methods: The model of acute reperfusion injury after cere-bral ischemia in rat was made by middle cerebral artery occlusion. All animals were allowed 45 min ischemia and sacrificedafter different durations of reperfusion (1, 7, 14, 21 and 28 days). The expression of MPA2 GFAP mRNA and protein aroundthe infarct area in the rat brain, and the effect of PTS on it were observed by PCR and western Results: The mRNA andprotein expression of MPA2 in the whole durations, the PTS groups was higher than that of normal sham and N.S groups (P〉0.05). In contrast, the mRNA and protein expression of GFAP increased remarkably as compared with the normal sham andN.S groups in 1, 7, 14, 21 and 28 days after reperfusion (P〈0.05). Conclusion: IriS can increase the expression of GFAP.PTS could enhance the level of NSC proliferation after middle cerebral artery occlusion reperfusion.
