摘要 目的探讨低分子量硫酸软骨素(CS)修复兔软骨的作用。方法 36只成年新西兰兔,随机分成6组,即对照组、模型组、低分子量CS低剂量组、低分子量CS高剂量组、高分子量CS低剂量组和高分子量CS高剂量组,每组6只。在实验兔股骨髁关节面部位钻直径3 mm,深3 mm缺孔,致其关节软骨缺损。术后次日药物灌胃,一日1次,持续5周,5周后取材。肉眼大体观察软骨缺损情况,光镜观察缺损部位软骨修复情况。采用酶联免疫法测定关节液中白细胞介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)的含量。结果与模型组相比,低分子量CS和高分子量CS均能明显促进兔软骨修复和伤口愈合,低分子量CS修复效果优于高分子量CS;低分子量CS和高分子量CS都能减少关节液中炎性因子的含量,与模型组相比差异有统计学意义(P<0.05);且低分子量CS优于高分子量CS(P<0.05)。结论低分子量CS能更好地促进伤口愈合和软骨修复,提高生物利用度;CS也可通过对IL-1β、TNF-α的影响,起到修复软骨的作用。 Objective To investigate the effect of chondroitin sulfate(CS) with low molecular weight on cartilage repair of rabbit.Methods Thirty-six New Zealand white rabbits were randomly divided into 6 groups with 6 rabbits in each groups:control group,model group,low-dose group of low molecular weight CS,high-dose group of low molecular weight CS,low-dose group of high molecular weight CS and high-dose group of high molecular weight CS.A defect(3mm diameter × 3mm depth) was drilled at the condyles of femur of the hind legs using an electric bone drill.From the next day after operation,drug perfusion was done once a day for 5 weeks.Then the cartilage was collected to observe the situation of cartilage defect.The light microscope was used to investigate the morphological change of cartilage.The expression of cytokines in synovial fluid,such as IL-1β and TNF-α,was detected by ELISA.Results Compared with the model group,low molecular weight CS and high molecular weight CS can significantly promote rabbit cartilage repair and wound healing,and the effect of low molecular weight CS was better than that of high molecular weight CS.Both of them can reduce levels of inflammatory cytokines in synovial fluid,and compared with the model group,the difference was of statistical significance(P<0.05).And the effect of low molecular weight CS was better than that of high molecular weight CS(P<0.05).Conclusion Low molecular weight CS can significantly promote rabbit cartilage repair and wound healing,and improve bioavailability.At the same time,CS can also affect the IL-1β and TNF-α to repair cartilage.
