摘要
[目的]观察鞘内注射Mas-related G Protein-coupled C(MrgC)抗体对电针(Electroacupuncture,EA)干预完全福氏佐剂(Complete?Freund's?Adjuvant,CFA)所致大鼠慢性炎性痛及脊髓背角(Spinal dorsal horn,SDH)N-甲基-D-天门冬氨酸受体亚基1(N-methyl-d-aspartate receptors1,NR1)表达的影响。[方法]SD雄性大鼠28只,脊髓鞘内插管成功后建立CFA模型,造模后随机分为生理盐水(Saline)组、MrgC抗体(MrgC Ab)组、MrgC Ab+EA组和Saline+EA组,每组7只。两EA干预组从造模24h后在患侧"足三里"和"昆仑"穴开始电针刺激,参数为2/100Hz、30min,1次/d,连续7d,其余各组均做相应固定处理。造模后7d,各组大鼠鞘内注射相应试剂。在造模前和造模后24h、6d、7d检测大鼠患足热缩退潜伏期(Thermal Paw Withdrawal Latency,T-PWL)。免疫组织化学法检测各组大鼠患侧SDH NR1的表达。[结果](1)各组大鼠在CFA造模前和造模后24h时,T-PWL均无明显差异。第6d时,与Saline组相比,MrgC Ab+EA组和Saline+EA组大鼠患足T-PWL显著延长(P<0.01),MrgC Ab组大鼠患足T-PWL无统计学差异。第7d时,与Saline组相比,MrgC Ab组大鼠T-PWL明显缩短(P<0.05),而Saline+EA组显著延长(P<0.01);与Saline+EA组比较,MrgC Ab+EA组大鼠T-PWL显著缩短(P<0.05)。(2)与Saline组比较,MrgC Ab组SDH NR1表达量显著提高(P<0.01);EA干预后大鼠SDH NR1表达量显著降低(P<0.01),MrgC Ab+EA组大鼠SDHNR1表达量也显著高于Saline+EA组大鼠(P<0.05)。[结论]鞘内注射MrgC抗体能显著拮抗电针对CFA诱导性慢性炎性痛的镇痛作用,电针镇痛效应可能与下调大鼠患侧脊髓背角NR1受体表达有关。
[Objective] To observe whether the intrathecal injection has an impact on the effect of electroacupuncture(EA) intervening on chronic inflammatory pain of rats that caused by complete Freund&#39;s adjuvant(CFA) and on the expression of spinal dorsal horn N-methyl-d-aspartate sub unit1 (NR1). [Methods] SD rats 28, each right hindfoot of rats was modeled with CFA after the success of the spinal cord sheath intubation.Al rats were randomly divided into four groups:saline,MrgC antibody ,MrgC antibody+EA and saline+EA. Each group of seven.Detect the thermol paw withdrawal latency(T-PWL) of rats 24h later,then startto intervene on the groups that EA is included at two points of affected side(&quot;Zusanli&quot; and &quot;kunlun&quot;), 2/100 Hz, 30 min, once a day and continuously for 7 days. Meanwhile, the other groups treated the same except for EA. On the 7d,each group was intrathecal injected of corresponding reagent.We tested the T-PWL of rats before the CFA modeled, 24h after modeled, the 6d and the 7d. The expression of NR1 in the spinal dorsal horn of each rats was detected by the method of immune histochemical. [Results] (1)The T-PWL of rats detected before CFA modeling and after modeling for 24h showed that each group was not statistical y different.The 6d, compared with Saline group, the T-PWL of MrgCAb +EA group and Saline + EA group rats were significantly prolonged( P〈0.01,P〈0.01), while the MrgCAb group was not statistical y different. The 7d, compared with Saline, the T-PWL of MrgC Ab group was significantly shortened but Saline+EA group was significantly prolonged( P〈0.05, P〈0.01);The T-PWL of MrgC Ab+EA group rats were no shorter than the Saline group, but significantly shorter than Saline+EA group .(2)Compared with Saline group, the expression of SDH NR1 was significantly increased in MrgC Ab group while decreased after intervened by EA( P〈0.01). Compared with Saline+EA group, the expression of SDH NR1 was significantly improved in the MrgCAb+EAgroup. [Conclusion]The analgesic effect of electroacu-puncture on chronicinflammatory pain induced by CFA would be strongly inhibited by intrathecal injectionof MrgC antibody,which may play a role in downregulating the expression of NR1 receptor distributed in the affected side spinal dorsal horn of rats.
