复方蜥蜴散不同微粒组合剂对胃癌前病变大鼠Stat3通路Stat3、Bcl-2蛋白表达影响的实验研究 被引量:14

Experimental study on the effects of Stat3 pathway and the expression of Stat3,Bcl-2 protein with the compound Lizard Powder on precancerous leision of gastric cancer in rats

摘要 目的研究复方蜥蜴散不同微粒组合剂对胃癌前病变大鼠Stat3通路Stat3、Bcl-2蛋白表达的影响,探讨复方蜥蜴散不同微粒组合剂对胃癌前病变抗凋亡蛋白的调控机制。方法将120只SD雄性大鼠随机分成空白对照组、复方蜥蜴散不同微粒组合剂80目治疗组、100目治疗组、80目和100目等量混合治疗组、胃癌前病变模型对照组、维酶素治疗组,除空白对照组外,均用N-甲基-N'-硝基-N-亚硝基胍、饥饱失常及情绪刺激综合因素8周制备胃癌前病变大鼠模型。分别用复方蜥蜴散不同微粒组合剂80目、100目、80目100目等量混合糊剂混悬液、生理盐水、维酶素治疗,观察大鼠胃黏膜病理组织学变化及应用免疫组化法检测胃体组织Stat3、Bcl-2蛋白表达情况。结果通过Image-Pro Plus 6.0图像分析系统对免疫组化图像进行光密度的测算,复方蜥蜴散不同微粒组合剂80目治疗组、100目治疗组、80目和100目等量混合治疗组Stat3、Bcl-2蛋白阳性表达率低于维霉素治疗组(P<0.05),其中以复方蜥蜴散80目100目等量混合治疗组差异最显著(P<0.01)。结论复方蜥蜴散不同微粒组合剂能部分逆转胃黏膜病理变化,降低Stat3、Bcl-2蛋白的表达,促进细胞凋亡,抑制细胞过度增殖,恢复细胞增殖与凋亡的平衡,揭示了复方蜥蜴散不同微粒组合剂可能是通过对抗凋亡蛋白的调控实现治疗胃癌前病变的作用机制的。 Objective To study the compound Lizards powder different particle combination agent on rats with precancerous lesion of gastric cancer Star3, BCL - 2 Protein expression of Star3 Pathway , to explore lizard compound combinations of different particles on the regulatory mechanism of PLGC of anti - apoptotic proteins. Methods 120 male SD rats were randomly divided into blank control group, 80 mesh, 100 mesh treatment group and 80 mesh and 100 mesh mixing equal treatment group,PLGC in model con- trol group, Vitacoenzyme treatment groups, except for the blank control group, with MNNC,irregular diet and emotional stimulation factor model of PLGC of rats 8 weeks. Respectively use 80 mesh, 100 mesh,80 mesh and 100 mesh mixing equal Paste suspension, physiological saline, Vitacoenzyme treatment, observation of rat gastric mucosa pathological changes and immunohistochemical method was used to detect the Stat3, Bel - 2 Protein expression of gastric tissue. Results Through the analysis of the Image - the Image Pro Plus 6.0 system for immunohistochemic^d Image optical density, the compound lizards powder different particle combi- nation agent 80 mesh, 100 mesh treatment group ,80 mesh and 100 mesh mixing equal treatment group, the Stat3, Bcl -2 Protein positive expression rate is lower than Vitacoenzyme treatment group (P 〈 0.05), in which the compound lizards powder different particle combination agent 80 mesh 100 mesh mixing equal is the most significant difference in treatment group ( P 〈 0.01 ). Con- clusion Compound lizards powder different particle combination agent can partly reverse gastric mucosa pathological changes, re- duce the expression of StaG, Bcl - 2 Protein, promote cell apoptosis, inhibit cell excessive proliferation and restore the balance of cell proliferation and apoptosis,and reveals the different complex lizards powder particle combination agent might be achieved by against the regulation of apootosis protein treatment on the mechanism of action of PLGC.
