丹参提取物对大鼠心肌梗塞后心肌组织VEGF表达的影响 被引量:8

摘要 目的探讨丹参提取物对大鼠心肌梗塞后心肌组织血管内皮生长因子(VEGF)表达的影响及其可能作用机制。方法建立大鼠心梗模型后,随机分为模型组、丹参提取物3个不同剂量组,每组8只大鼠,另设假手术组8只。术后48 h,丹参提取物组分别给予低、中、高10,20,40 mg·(kg·d)-1灌胃;模型组和假手术组给予生理盐水20 ml·(kg·d)-1灌胃。8周后处死大鼠,取大鼠心肌组织,反转录PCR(RT-PCR)法测定VEGF mRNA的表达。应用免疫组化和免疫印迹法分析左心室心肌组织VEGF蛋白的表达情况。结果 RT-PCR结果表明,和模型组相比,丹参提取物各剂量组VEGF mRNA的表达均明显升高(P<0.01)。免疫组化和免疫印迹分析结果表明,和模型组相比,丹参提取物各剂量组心肌组织胞浆中VEGF蛋白的表达均显著升高(P<0.01)。结论丹参提取物可以通过上调VEGF的表达而促大鼠心肌梗塞后心肌组织的血管新生。 Objective To study the effect and mechanism of Salvia extract on the expression levels of VEGF in rats with myocar- dial infarction (MI). Methods Left coronary artery of Sprague -Dawley(SD) rats were ligated to make MI models. The rats were randomly subjected to MI model group, sham -operated group, and three different salvia extract (SE) (10,20,40 mg · (kg·d) -1) groups,each group consisted of 8 rats. The MI group and shamoperated group were fed O. 9% sodium chloride 20 ml · ( kg·d) -1.8 weeks later, the rats were sacrificed, the segmental heart samples were used for histological evaluation on expression of vascular endothelial growth factor(VEGF). Immunobloting analysis was also employed to determine alterations in protein expression of VEGF. And mRNA expression of VEGF was employed to determine by the reverse transcription PCR (RT -PCR) method. Results Compared with the model group, total ~EGF mRNA expression were significantly increased ( P 〈 0.05 ) in the SE groups. Compared with the model group, VEGF protein expression in the cytoplasm of myocardial tissue of the SE group increased signifi- cantly ( P 〈 0.05 ) by histological evaluation, and the immunobloting analysis the same results. Conclusion Salvia extract can sig- nificantly increase the expression of VEGF,which is probably related to angiogenesis of myocardial tissue in rats with myocardial infarction.
