摘要
目的建立超声波热水浸提法提取油松松子壳多糖最佳工艺条件。方法利用超声波热水浸提法,苯酚-浓硫酸法检测多糖含量,选择浸提温度、时间、固液比和浸提次数进行单因素梯度实验,再通过L9(34)正交实验优选出油松松子壳多糖提取最佳条件。结果与结论最佳提取条件为浸提温度为60℃,时间为25 min,固液比为1∶3.5,反复浸提2次。该方法所得回收率(n=5)在97.71%~100.67%之间,RSD为1.56%,稳定性较好,为废弃松子壳资源的开发利用建立了一个简单高效的提取方法。
Objective This paper studied the uhrasonic hot water extraction of the polysaccharide from Pinus seed shell. Methods The effects of four factors such as the temperature of extraction, the time of extraction, the proportion of water volume, the number of extraction on the extraction were investigated. Results and Conclusion With the orthogonal design of L9 ( 34 ), the optimum conditimls of extraction were obtained as follows:temperature 60~C ,Time 25 min, ratio of solid - liquid 1:3.5, extraction twice. Tests for recovery were made by standard addition method, values of recovery found ( n = 5 ) were in the range of 97.71% - 100. 67% and value of RSD was 1.56%. Based on these factors, a easy and efficient method of sepectrophotometric determination of polysaccharide in Pinus seed shell with ultrasonic extraction.
