摘要
目的研究在人工胃液和人工肠液环境下马钱子与地龙不同配伍比例前后对生物碱成分士的宁和马钱子碱含量的影响。方法采用HPLC法,色谱柱:大连依利特(Hypersil ODS 25μm,4.6mm×250 mm);流动相:乙腈(A)∶水-乙酸-三乙胺(230∶2.4∶0.4)(B)=11∶89;检测波长:258 nm;柱温:30℃;流速:1.0ml/min。结果马钱子配伍地龙后,在人工胃液环境中,士的宁、马钱子碱含量表现为总体上降低的趋势,其中马钱子地龙1∶1配伍组含量最低。在人工肠液环境中,士的宁、马钱子碱含量显著降低。人工肠液对士的宁、马钱子碱的影响大于人工胃液。结论马钱子与地龙配伍后,模拟人工胃肠液条件,其有毒成分士的宁、马钱子碱含量都减少,可以达到减毒的目的。研究为扩大马钱子的临床应用提供了科学的依据。
Objective To do research on the content of Strychnine and Brucine in Strychnos nux -vomical L. Uncombined and Combined with earthworm by artificial gastric intestinal fluid environment. Methods The chromatographic conditions were as follows : Hypersil ODS 25 μm 4.6 mm×250 mm, mobile phase : acetonitrile (A) : water - acetic acid - triethylamine ( 230:2.4:0. 4) (B) = 11:89, detection wavelength at 258nm, the column temperature at 30℃ and flow phase rate being 1.0mL/min. Results Combined with earthworm, by artificial gastric fluid environment, the content of Strychnine and Brucine totally has the trend of Re- ducing. Compatibility group 1 : 1 has the lowest content. By artificial intestinal fluid environment, the content of Strychnine and Bru- cine totally has the significantly reduced. The effect in the artificial intestinal is greater than the impact in artificial gastric juice on the strychnine and brucine. Conclusion Combined with earthworm, under the condition of artificial gastrointestinal fluid, the con- tent of toxic components strychnine and brucine are reduced. It can achieve the purpose of attenuated. It provides a scientific basis on expanding the clinical application of Strychnos.
