摘要
目的:探讨河南省间日疟原虫环子孢子蛋白( CSP )的基因型分布。方法:收集经镜检确诊的河南省2011年间日疟患者血标本34份,24例患者为河南籍,发病前无外出到其他省份或国家的历史,10例患者有明确外出史。用核酸提取试剂盒提取滤纸血中疟原虫DNA,进行巢式PCR扩增,对扩增产物进行序列测定和基因进化特征分析。结果:24份本地病例分离所得的本地株间日疟原虫CSP均属于PV-Ⅰ型,又可分为A和B 2个基因型;氨基酸同源性分析结果显示,A型与VK210的同源性达90.6%~94.2%,B型为71.6%。结论:河南省间日疟原虫CSP的基因型均属PV-Ⅰ型,可以进一步分为2个基因型,与其他国家的基因型显著不同。
Aim:To explore circumsporozoite protein (CSP) genotypes structure of Plasmodium vivax(P.vivax) in Henan.Methods:Blood samples of 34 malaria patients from Henan Province in 2011 were collected to extract DNA ,and amplified using nested PCR technique , and the product was sequenced and analyzed .Results:The results showed that the 24 specimens from Henan indigenous cases all belonged to PV-Ⅰ;amino acid homology analysis showed that the specimens could be divided into two genotypes:A and B, the sequence of type A showed 90.6%-94.2%homology to VK210, while type B showed 71 .6%.Conclusion:The P.vivax CSP genotype of Henan all belongs to PV-Ⅰ, and it can be further di-vided into two genotypes , which is significantly different from other countries .
