摘要
采用超声辅助提取,建立甜叶菊中A苷甜菊糖和甜菊苷的高效液相色谱分析方法。采用Kroma-sil C18(4.6mm×250mm,5μm)色谱柱,以甲醇:水(75:25,V/V)为流动相,流速1.0mL/min,检测波长210nm,柱温30℃。A苷甜菊糖进样量在0.868~8.68μg时,与峰面积呈良好的线性关系(R=0.999),平均回收率为97.5%,RSD为1.8%(n=6);甜菊苷进样量在1.004-10.04μg时,与峰面积呈良好的线性关系(R=0.999),平均回收率为95.6%,RSD为1.6%(n=6)。该方法准确、操作简便,可用于甜叶菊中A苷甜菊糖和甜菊苷含量测定。
HPLC method for the determination of rebaudioside A and stevioside in Stevia rebaudiana Bertoni is established by ultrasonic assisted extraction method. The separation is achieved on a Kromasil C18 column (4.6 mm×250 mm, 5 μm) at 30 ℃ using methanol and water solution (75 : 25, V/V) as the mobile phase at a flow rate of 1.0 mL/min, 210 nm as the detection wavelength. Rebaudioside A shows good relationship at the range of 0. 868-8.68 μg (R=0.999), the recovery is 97.5% and RSD is 1.8% (n=6). Stevioside shows a good relationship at the range of 1. 004-10.04 μg (R= 0. 999), the recovery is 95.6% and RSD is 1.6% (n=6). The proposed method is accurate, simple and applicable for the determination of rebaudioside A and stevioside in Stevia rebaudiana Berton.
