摘要
黑曲霉2是本实验筛选的一株具有降解有机废弃物的功能微生物,为进一步深入研究其产酶条件,提高应用效果,本研究基于现有研究结果,采用液体发酵培养法,分别测定了培养时间、接种菌龄、通气状况、初始pH、不同碳源底物对其产酶活性的影响。实验结果表明:黑曲霉2在采用原始菌株接种、基础培养基初始pH为6.5、每500mL摇瓶装量为150mL、发酵48h条件下产纤维素酶活性相对较高;同时采用不同碳源底物影响产纤维素酶活性,活性从高到低依次为木薯渣、中药渣、稻秸和糖泥。
Aspergillus niger 2 is a functional microbe used for degradation of organic waste which was isolated by college of environmental science and technology,Zhongkai University of Agriculture and Engineering.Based on present research,different conditions such as culturing time,inoculating age,ventilation,initial pH and carbon source substrate were tested on activity of cellulase for Aspergillus niger 2 by liquid-state fermentation.The results indicated that the activity of cellulase was relativity higher under the conditions of inoculating the original strain,adjusting the initial pH 6.5 for basic medium,loading 150mL media per 500mL bottle and culturing for 48h.Meanwhile,the activity of cellulase was diverse when adopting different carbon source substrate,and the activity from high to low as below:cassava dregs,Chinese medicine residue,rice straw and sugar mud.
