Quick Method for Nicarbazin Determination in Feedstuffs Using HPLC- MSD

建立了简单、快速、灵敏、节约的饲料中尼卡巴嗪含量的高效液相色谱色谱一单极质谱检测方法。分析物经二甲基亚砜（DMSO）-乙腈（ACN）（4：6，体积比）混合溶剂提取，氯化钠、硫酸镁和C18粉末净化，冷冻高速离心。采用梯度洗脱。外标法定量。在饲料中添加0．5、1．0、1．5mg／kg尼卡巴嗪进行重复性试验，结果表明，尼卡巴嗪回收率在93．7％。104．9％之间，相对标准偏差（RSD）不大于3．9％（n=6）。饲料中其它常用的抗球虫药如地克珠利、妥曲珠利及其代谢物等对该检测无干扰。

A simple, rapid,sensitive and cheaper HPLC-MSD method had been developed for the determination of nicarbazin （4,4＇-dintrocarbanilide, DNC） in feedstuffs. The analytes were extracted by dimethylsulfoxide （DMSO） -acetonitrile （4 ： 6, v/v） and cleaned up by sodium chloride, magnesium sulfate and C18 powder. The quantification was performed with the external standard method. The recoveries of DNC were within 93.7 % and 104.9 % with spiked level of 0.5, 1.0, 1.5 mg/kg, the RSD was not more than 3.9 %（n=6）. There was no interference from other Anticoccidials drugs in feeds such as diclazuril, tohrazuril and its metabolites in the determination of nicarbazin.
