摘要
目的 研究鬼臼酰肼哌啶氮氧自由基腙 (GP1)和阿霉素 (DOX)协同抗肿瘤作用。方法 用MTT比色法观察GP1、DOX对K56 2细胞生长的抑制作用。结果 GP1和DOX单独应用时 ,最大浓度 (10 μg·ml- 1)对K56 2细胞的抑制率分别为 2 0 5%和 4 6 0 % ,当用GP11μg·ml- 1和不同浓度的DOX联合应用时 ,对K56 2细胞的抑制作用明显增强 ,DOX的IC50 为 3 87μg·ml- 1,是单用DOX(IC50 为 14 32 μg·ml- 1)的 1/3 7。联合用药的抑制率大于单独用药的总和 ,q >1 15。结论 GP1和DOX两药合用有协同抗肿瘤作用。
Objective To study the cooperative antitumor e ffects of podophyllic acid[4-(2,2,6,6-tetramethy1-piperidinooxyl)] hydrazone(GP1)and doxorubicin(DOX).Methods The effects of GP1 and DOX on K562 cell line were studied by MTT colorimetry.Results The inhibition rates of 10 μg·ml -1 GP1 and DOX to K562 cells were 20 5% and 46 0% respectively when given alone,but they were greatly increased when GP1 1 μg·ml -1 was used with D OX at different concentrations.IC 50 of DOX (3 87 μg·ml -1 )was 1/ 3 7 times than that of DOX(14 32 μg·ml -1 )given alone.The inhibition ra tes of GP1 combined with DOX were greater than the total inhibition rates of sin gle drug,q>1 15 Conclusions GP1 and DOX have co operative antitumor effects.
