摘要
目的:筛选并确定维吾尔药宝心艾维西木口服液中可用于其质量控制的含量测定指标。方法:利用液相色谱.串联质谱(LC—MS/MS)法进行分析测定。色谱柱为Waters 5C18-PAQ18(250mm×4.6mm,5μm),流动相为乙腈-0.2%乙酸水溶液(梯度洗脱),流速为1.0ml/min,离子源为ESI,采用正负离子扫描模式,温度为400℃,质量扫描范围(m/z)为100~1000。结果:共鉴定出7个组分,分别推测为咖啡酸、麝香草酚、山柰酚-3-O-d-(6-O-对羟基桂皮酰)半乳吡喃糖苷、芹菜素-7-O-β-d-移小葡萄糖、射干苷、迷迭香酸、田蓟苷;其中迷迭香酸为主要成分,故将其选定为该制剂质量控制中的含量测定指标。结论:利用LC—MS/MS法对维药中的复杂成分进行分析,快速获得化合物的结构信息,及时识别已知化合物,可快速筛选出合适的含量测定指标。
OBJECTIVE: To select and determine the index of content determination of quality control of Baoxin aiweiximu oral liquid. METHODS: LC-MS/MS method was used. The determination was performed on Waters 5C18-PAQ(250 mm×4.6 mm, 5 μm) column with mobile phase consisted of acetonitrile-acetic acid (gradient elution) at the flow rate of 1.0 ml/min. ESI was applied and operated in both negative and positive modes at 400 ℃, full scan (m/z) from 100-1000 amu. RESULTS: 7 compounds were identified, including caffeic acid, thymol, kaempferol-3-O-d-(6-O-p-hydroxy cinnamyl) galactopyranose, apigenin-7-O- β-d-glucose, belamcandin, rosmarinci acid and tilianin; as main component, rosmarinic acid was selected as the index for quality control of the preparation. CONCLUSIONS: LC-MS/MS is adopted to analyze the comprehensive components of Uyghur medicine to quickly obtain the information of compound structure and screen suitable content determination indexes.
