摘要
[目的]探讨重组生长激素对梗阻性黄疸大鼠肠黏膜屏障的保护作用及血清转化生长因子β1(TGF-β1)水平的影响.[方法]将Wistar大鼠随机分为假手术(SO)组、梗阻性黄疸(OJ)组、生长激素(rhGH)组.每组术后7,14 d检测血清总胆红素(TBIL)、直接胆红素(DBIL)和谷丙转氨酶(ALT)值,检测大鼠门静脉血浆内毒素TGF-β1含量.[结果]OJ组和rhGH组血浆内毒素水平明显高于SO组(P<0.05);且随着梗阻时间的延长逐渐增高(P<0.05);rhGH组血浆内毒素水平与OJ组比较明显降低(P<0.05).OJ组血清TGF-β1水平显著高于SO组(P<0.05);rhGH组血清TGF-β1水平明显低于OJ组(P<0.05).[结论]外源性生长激素可改善梗阻性黄疸大鼠肠黏膜屏障功能,降低血浆内毒素水平.血清TGF-β1水平在一定程度上反映肠黏膜屏障功能的损害.
OBJECTIVE To study the effects of function of intestinal mucosal barrier and TGF-β1 recombinant human growth hormone(rhGH) on protective in rats with obstructive jaundice. METHODS Wastar rats were randomly divided into sham operation(SO), obstructive jaundice(OJ) and rhGtt groups, and it was measured in each group after operation at 7, 14 days that contents of total bilirubin(TBIL), direct bilirubin (DBIL), alanine aminotransferase(ALT) in blood and endotoxin and transforming growth factor-β1 (TGF-β1) in serum of portal vein. RESULTS The content of endotoxin was higher in OJ and rhGH groups than in SO group(P〈0.05), and was gradually increased along with extending time of OJ(P〈0.05), and was lower in rhGH group than in OJ group(P〈0.05). The content of TGF-β1 was higher in OJ group than in SO group (P〈0.05), and was lower in rhGH group than in OJ group(P〈0.05), than those in the group of SO (P〈 0.05); Blood TGF-β1 in the groups of rhGH were less than those in the group of OJ (P 〈 0.05). CONCLUSION Ectogenic growth hormone that could improved intestine mucosal barrier function and could depressed endotoxin level in blood, the level of blood TGF-β1 could reflected damage of intestine mucosal barrier function.
