摘要
考察了培养基组成 ,培养条件对产朊假丝酵母生长和谷胱甘肽合成的影响 ,包括不同碳源 ,氮源及其浓度 ,p H,装液量 ,接种量等。确定以葡萄糖、磷酸氢二铵作为碳源和氮源 ,较佳浓度分别为 2 0 g/L和 3g/L ,接种量为 10 %。在 2 0 g/L的糖浓时获得的谷胱甘肽总量可达 74mg/L,胞内含量为 1.4%左右。此外还研究了三种前体氨基酸和 ATP的加入量 。
The effects of culture conditions and medium on production of glutathione by Candida utilis were investigated in shaking-flask culture, including different carbon source, nitrogen source and their concentrations, pH, working volume and inoculum. Glucose and (NH4)2HPO4 were determined with the optimal concentration of 20g/L and 3g/L respectively. The optimal inoculum was 10%. A total glutathione amount of 74mg/L was achieved with the initial glucose concentration of 20g/L. The intracellular GSH content was increased to 1.40% by changing the culture conditions. The effects of the three constituent amino acids of glutathione and ATP on GSH production were also studied.
