熊果酸对球囊损伤大鼠颈总动脉后内膜增生及PCNA、NF-kB表达的影响 被引量:4

摘要 目的探讨熊果酸对球囊损伤大鼠颈总动脉后血管内膜的增生及增殖细胞核抗原(PCNA)、核因子kB(NF-kB)表达的影响。方法 32只SD大鼠随机分为假损伤组(对照组)、损伤组和熊果酸低剂量治疗组、高剂量组治疗组。对照组和模型组每天腹腔注射生理盐水及药物助溶剂1%DMSO,熊果酸低剂量治疗组、高剂量组治疗组每天腹腔注射相应药物。分别在术后7 d、21 d取颈总动脉,通过组织学检查、免疫组化结合图象分析技术测定血管壁形态学变化及PCNA、NF-kB的表达情况。结果与损伤组相比,术后21天熊果酸高、低剂量组内膜增生面积及内膜与中膜面积比均明显减少(P<0.05)。与对照组比较,损伤组NF-kB、PCNA表达显著增强(P<0.05);术后各时间点上熊果酸高、低剂量组较损伤组NF-KB表达明显降低,差异有统计学意义(P<0.05)。结论熊果酸可抑制球囊损伤后血管新生内膜增生,其作用机制可能与抑制NF-kB及PCNA表达有关。 Objectiveto:Investigate the effect of ursolic acid on neointimal hyperplasia and expression of PCNA、NF-kB af-ter aortic balloon injury. Methods Thirty-two healthy male SD rats were randomly divided into the sham-operated,the in-jury group,ursolic acid 10mg/kg intervention group,ursolic acid 20mg/kg intervention group. Sham-group and injury-group that received equal volume PBS including1%DMSO;ursolic acid 10mg/kg and ursolic acid 20 mg/kg received corre-sponoding ursolic acid everyday. Carotid tissues of four groups were harvested 7、21day after Carotid artery injury. The target vascular sections were stained with HE to observe the morphology changes. The expression of NF-kB and PCNA in arterial wall was detectde by immunohistochemistry. Result:both neointimal area and IA/MA were significantly reduced in ursolic acid group as compared to that in injury group at 21 day points after balloon injury(P﹤0. 05). Compared with the sham-group,the expression of NF-kB and PCNA positive cells were significantly increased in the injury group(P﹤0. 05). Com-pared with the injury-group,The expression of NF-kB were significantly decreased in ursolic acid group at different time points after balloon injury(P﹤0. 05). Conclusions:ursolic acid can inhibits neointimal hyperplasia after balloon injury. The possible mechanism is connected with inhibition the expression of NF-kB and PCNA.
