摘要 目的:采用逆流萃取的方法,在两种不同的萃取流速下,考察两种不同产地栀子的栀子苷含量及稳定性,以期对栀子药材的粗提工艺提供一定的参考。方法:采用HPLC法测定不同栀子逆流萃取液中栀子苷含量。以十八烷基硅烷键合硅胶为填充剂C18色谱柱(5μm,4.6×250 mm);流动相乙腈一水(15∶85);流速1.0 mL·min-1;检测波长为238 nm;柱温30℃。结果:两种不同产地不同逆流萃取条件下,栀子苷均在5 h达到最大值,种植栀子比野栀子中栀子苷含量高,且同一产地栀子在B流速时大于A流速时栀子苷含量,1个月后重复测定1次,栀子苷含量稳定。结论:HPLC简便快速、灵敏度高、重复性好、稳定性好,可为栀子提取工艺研究提供参考。 Objective : Using the method of reflux extraction, under two different extraction velocities to determinate and investigate two different origin gardenia extracting conditions of geniposide contenta and stability, in order to provide certain reference for coarse extraction technology of gardenia medicinal materials. Methods: HPLC method was used for the determination of geniposide in different gardenia extraction velocity solutions : octadecyl silane bonded silica as filler C18 column(5 μm,4.6 mm ×250 mm);acetonitrile-water (15:85) as mobile phase;flow rate 1.0 mL · min^-1 ;detection wavelength 238 nm and column temperature 30℃. Results : Two different conditions of different counter - current extractions and geniposide in five hours reached the maximum. The planting gardenia' s geniposide content was higher than the wild gardenia geniposide' s and for the same origin gardenia, the geniposide content was greater under B velocity than that under A velocity. Repeat measurement in a month time showed the geniposide content was stable. Conclusion:This established HPLC was convenrent and accurate with good repeatability and good stability. It can provide reference for gardenia extract technology.
