摘要
目的:分析苦参注射液与卡铂联合应用对人原发性肝癌细胞凋亡的协同增效作用。方法:以HEP肝癌细胞作为Ⅰ组,SMMC-7721肝癌细胞作为Ⅱ组。采用原位末端标记法(TUNEL)观察此化疗方案对肿瘤细胞凋亡的影响;采用MTT比色法观察其对肿瘤细胞增殖的影响;应用流式细胞术(FACS)分析其对肿瘤细胞周期的影响。结果:苦参注射液凋亡指数(AI)分别为(23.6±2.4)%(Ⅰ组)、(24.3±3.3)%(Ⅱ组),卡铂的AI分别为(32.2±3.9)%(Ⅰ组)、(29.7±3.2)%(Ⅱ组)。两者联合AI分别为(41.5±3.6)%(Ⅰ组)、(36.8±4.1)%(Ⅱ组)(P值均<0.05)。苦参注射液抑瘤率(IR)分别为(41.1±4.3)%(Ⅰ组)、(39.9±5.3)%(Ⅱ组),卡铂的IR分别为(48.9±4.9)%(Ⅰ组)、(45.7±5.8)%(Ⅱ组)。两者联合IR分别为(65.2±5.8)%(Ⅰ组)、(54.1±6.3)%(Ⅱ组)(P值均<0.05)。苦参注射液阻滞于G0/G1期的比例分别为(40.8±3.5)%(Ⅰ组)、(38.2±4.9)%(Ⅱ组),卡铂比例分别为(53.4±4.3)%(Ⅰ组)、(51.1±5.2)%(Ⅱ组)。两者联合的比例分别为(72.4±4.7)%(Ⅰ组)、(68.2±5.6)%(Ⅱ组)(P值均<0.05)。结论:苦参注射液与卡铂联合应用能更加有效诱导肝癌细胞株的凋亡。
Objective: To investigate the synergistic effect of Kushen Injection combined with Carboplatin for apoptotic action on human primary hepatoma cell lines. Methods: The hepatoma cell HEP was chosen to Ⅰ group and SMMC - 7721 to Ⅱ group. The TUNEL method was employed to observe the cell apoptosis applied by Kushen Injection combined with Carboplatin. The MTT test was used to estimate the inhibition of proliferation and flow cytometry (FACS) was applied to analyze the distribution of cell cycle. Results: The apoptotic indexes (AI) by applying Kushen Injection alone were (23.6± 2.4) % ( Ⅰ group) vs (24.3 ± 3.3 ) % ( Ⅱ group) respectively ( P 〈 0.05 ). The AI by applying Carbopl atin alone was (32.2 ±3.9) % ( Ⅰ group) vs (29.7 ±3.2)% ( Ⅱ group) respectively (P〈0.05). The AI by applying Kushen Injection combined with Carboplatin was (41.5 ± 3.6) % ( Ⅰ group) vs ( 36.8 ± 4.1 ) % ( Ⅱgroup) respectively ( P 〈 0.05 ). The inhibitory rate (IR) by applying Kushen injection alone was (41.1 ± 4.3 ) % ( Ⅰ group) vs (39.9 ± 5.3 ) % ( Ⅱ group) respectively (P 〈 0.05 ). The IR by applying Carboplatin alone was (48.9 ± 4.9) % ( Ⅰ group) vs (45.7 ± 5.8) % ( Ⅱ group) respectively (P 〈 0.05). The IR by applying Kushen Injection combined with Carboplatin was (65.2 ± 5.8 ) % ( Ⅰ group) vs ( 54.1 ± 6.3 ) % ( Ⅱ group) respectively ( P 〈 0.05 ). The ratios of cells blocked at G0/G1 phase by applying Kushen Injection alone were (40.8 ± 3.5 ) % ( Ⅰ group) vs (38.2 ± 4.9 ) % ( Ⅱ group) respectively (P〈0.05). The ratios by applying Carboplatin alone were (53.4 ±4.3) % (Ⅰgroup) vs (51.1 ± 5.2)% ( Ⅱ group) respectively (P 〈 0.05 ). The ratios by applying Kushen Injection combined with Carboplatin were (72.4 ± 4.7 ) % ( Ⅰ group) vs (68.2 ± 5.6) % ( Ⅱ group) respectively ( P 〈 0. 05 ). Conclusion : The synergistic effect of Kushen Injection combined with Carboplatin for apoptotic action on human primary hepatoma cell lines was perfect.
