摘要
目的研究西黄丸乙酸乙酯提取物抗肿瘤作用及对荷瘤大鼠免疫功能的影响,筛查西黄丸的抗肿瘤活性部位。方法用W256乳腺癌细胞建立荷瘤大鼠模型,随机分为正常对照组,模型对照组,阳性药物(香菇多糖)组,西黄丸高剂量组,西黄丸乙酸乙酯提取物高、中、低剂量组。给药治疗14 d,取瘤称重,计算抑瘤率;腹主动脉取血,流式细胞仪检测外周血T淋巴细胞亚群及B7-1的含量;酶联免疫吸附(ELISA)法检测外周血白细胞介素2(IL-2)、IL-6、IL-10、β转化生长因子(TGF-β)、γ干扰素(IFN-γ)表达水平。结果西黄丸乙酸乙酯高剂量组抑瘤率30.41%;西黄丸乙酸乙酯提取物高剂量组CD+3、CD+8、B7-1含量及IL-2、IFN-γ水平均高于模型对照组,IL-6及TGF-β水平均低于模型组;CD+4、IL-10与阴性对照组比较,均差异无统计学意义(P>0.05)。结论西黄丸乙酸乙酯提取物通过上调IL-2、IFN-γ水平及提高CD+8细胞含量,降低肿瘤相关炎性因子IL-6、TGF-β水平,提高了机体免疫清除功能,发挥抗肿瘤作用,提高原药生物利用度。乙酸乙酯提取物可能是西黄丸的主要活性部位之一。
Objective This research aimed to study the antitumor activity and immunologic function of ethyl acetate ex-tracts from xihuang pills on tumor-bearing rats. Methods The Walker256 bearing rats were randomly divided into 7 groups (n =10): normal control,model control,different doses of ethyl acetate extracts from xihuang pills ,and lentinan plus high doseof xihuang pill groups. After the drug administration for a consecutive 14 days,the inhibitory rate of tumor was measured. FCMmethod was used in determination of serum levels of CD+3 、CD+4 、CD+8 、B7-1. ELISA method was applied in measuring serum lev-els of IL-2, IL-6, IL-10, TGF-β and IFN-γ. Results The ethyl acetate extract from xihuang pills effectively inhibited thegrowth of Walker256 by 30. 41%. Compared with the model control, serum levels of IL-2, IFN-γ and CD+3 、CD+8 、B7-1 were sig-nificantly higher (P〈0. 05),and those of IL-6 and TGF-β were significantly lower (P〈0. 05) in the high dose xihuang pill ethylacetate extracs group. No significant difference in the level of CD+4 and IL-10 was found in the control. Conclusion The ethylacetate extracts from xihuang pills is one of the active constituent parts for inhibition of tumor growth by elevating IL-2, IFN-γand CD+8 , B7-1 and reducing IL-6 and TGF-β.
