血浆中亚甲蓝释放量和残留量的超分子荧光分析 被引量:2

Fluorescence analysis of released and residual contents for methylene blue in plasma by using supramolecular complex

摘要 利用羧甲基-β-环糊精对亚甲蓝荧光信号的增敏作用,用荧光分光光度法直接测定血浆中的亚甲蓝的释放量和残留量。结果表明:亚甲蓝浓度在0.080~8.000μmol·L-1范围内呈现良好的线性关系,R2=0.9987。高中低浓度的日内CV分别为0.36%、0.85%和0.38%;日间CV分别为1.26%、1.56%和2.31%;回收率为102.02%~107.15%。通过此方法的检测,20个实际样品亚甲蓝释放量和20个实际样品亚甲蓝残留量均符合标准。本法准确、简便、快速,检测灵敏度高,可同时用于血浆中亚甲蓝的释放量和残留量的测定。 Based on the sensitizing effect of carboxym-ethyl-β-cyclodextrin on the fluorescence signal of methylene blue,the released and residual quantity of methylene blue in plasma was determined directly by the fluorescence spectrophotometry. Results indicated that when using carboxymethyl-β-cyclodextrin as the sensitizer, the concentration of methylene blue in 0. 080 ~8. 000μmol·L-1 showed a good linear relationship(R2=0. 9987),and the intraday coefficient variation(CV)of the high,medium and low concentra-tions were 0. 36%、0. 85%and 0. 38%,and the days CV of three samples were 1. 26%、1. 56%and 2. 31%,respectively,and the recovery rate was 102. 02% ~107. 15%. The released and residual quantity of methylene blue in plasma sample determined meet standards. The method is simple,accurate,fast,sensitive and could be used for the determination of the released and residual con-tent of methylene blue in plasma.
