摘要
为了克隆NALP3基因的部分序列及检测其在鸡体内各组织中的表达情况,本试验选择12周龄的公、母清远麻鸡各3只,分别采集其心脏、肝脏、脾脏、肺脏、肾脏、胸腺、法氏囊、小肠、输卵管、卵巢、睾丸和脑12种器官,通过RT-PCR(Reverse transcription PCR)技术克隆NALP3部分cDNA序列,并采用Real time-PCR方法检测NALP3 mRNA的表达水平。结果显示,获得了清远麻鸡NALP3部分cDNA序列,序列长度为210 bp。在心脏、肝脏、脾脏、肺脏、肾脏、胸腺、法氏囊、小肠、输卵管、卵巢、睾丸和脑中均有NALP3 mRNA表达,其中在心脏中表达量最高,在小肠中表达量最低。
To clone partial fragment of NALP3 gene and detect its expression levels in various tissues of chickens,the samples of heart, liver, spleen, lung, kidney, thymus, bursa, small intestine, fallopian tubes, ovaries, testicles andbrain were collected from 12-week-old Qingyuan partridge chickens, each three for male and female. NALP3 gene wascloned by reverse transcription PCR, and its expression levels in twelve organs of chickens were detected by real time-PCR.The results showed that partial cDNA sequence (210 bp)of NALP3 gene in Qingyuan partridge chickens was successfullycloned. The expressions of NALP3 mRNA varied in 12 organs, the highest level in heart, followed by thymus and the lowestin small intestine.
