摘要
目的 :探讨体外热应激诱导胸腺细胞凋亡与细胞表达HSP 70的关系。方法 :用流式细胞术检测体外热应激后再培养的胸腺细胞不同时间的凋亡率及HSP 70表达阳性率。结果 :体外热应激后 ,胸腺细胞的凋亡率在 0h、6h、12h随时间延长而增高 ,但 2 4h后凋亡率不再增高 ;体外热应激胸腺细胞HSP 70表达阳性率比对照组明显增高 ,在 6h阳性率最高 ,12h、2 4h阳性率逐渐下降 ,至 2 4h ,已明显低于 6hHSP 70的阳性率 (P <0 .0 5 )。结论 :HSP 70是调节体外热应激诱导胸腺细胞凋亡的重要分子之一。
Objective: To study the relationship between heat stress induced thymocyte apoptosis and the expression of HSP 70 in vitro. Methods: To measure the rate of thymocyte apoptosis at different time and the expression of HSP 70 by flow cytometry. Results: Apoptotic rate of heat stress induced thymocytes increased time dependently, i.e. markedly higher than those of the controls at 6 h and 12 h, but after 24 h,no more increase could be seen. The expression of HSP 70 in heat stress treated thymocytes was obviously higher than those of the controls, and reached the peak at 6 h, and decreased progressively at 12 h and 24 h. Conclusion: HSP 70 is one of the important molecules which regulate heat stress induced apoptosis of thymocytes in vitro.
