摘要
为了探讨金匮肾气丸水提液对肾阳虚雄鼠睾丸和肾上腺形态结构及功能的影响,试验将昆明小鼠分为空白对照组,模型组,阳性药物对照组和金匮肾气丸水提液高剂量组[20.30 g/(kg·d)]、中剂量组[13.50 g/(kg·d)]、低剂量组[6.08 g/(kg·d)],采用肌肉注射氢化可的松造模,灌服金匮肾气丸水提液进行治疗,试验期40 d,检测小鼠体质量增重率、肾上腺指数、睾丸指数及睾丸形态结构。结果表明:灌服金匮肾气丸水提液后,小鼠生长性能得到明显改善,睾丸指数和肾上腺指数增加,与模型组相比,差异显著或极显著(P<0.05或P<0.01或P<0.001),20.30 g/(kg·d)、13.50 g/(kg·d)金匮肾气丸水提液效果优于6.08 g/(kg·d),差异显著(P<0.05);H.E.染色镜检可见,药物组小鼠睾丸形态结构的病理变化明显好转,且剂量为20.3 g/(kg·d)效果最佳。说明金匮肾气丸治疗肾阳虚小鼠的机制可能与其保护睾丸和肾上腺有关。
To investigate the effects of water extract of Jinkuishenqi pills on the morphological structure and function of the testes and adrenal gland in male mice with kidney - yang deficiency, the Kunming mice were divided into six groups, namely the blank control group, model group, positive drug control group and three Jinkuishenqi pill groups including high dose group with 20.30 g/( kg · d), medium dose group with 13.50 g/( kg · d) , and low dose group with 6.08 g/( kg · d). The mice were injected intramuscularly with hydroeortisone to establish the model of the kidney - yang, and then were drenched with the three doses of water extract of Jinkuishenqi pills. The test lasted 40 d, and the changes in the rate of weight increase, adrenal indices, testicular indices, and morphological structure of the testes in the mice were deter- mined. The results showed that, after drenching with the water extract of Jinkuishenqi pills, there was a significant improvement on the growth performance of the mice, and the testicular indices and adrenal indices were increased; there were significant or extremely significant differences compared with the model group ( P 〈 0.05 or P 〈 0.01 or P 〈 0. 001 ). The effects of 20.30 g/( kg · d) and 13.50 g/( kg · d ) Jinkuishenqi pills were better than the effect of 6.08 g/( kg · d) ( P 〈 0.05 ) , there were significant differences among them. The result of H. E. staining showed that three Jinkuishenqi pill groups had obvious effects on treating the pathological changes in the morphological structure of the testes of mice, 20.30 g/( kg· d) of water extract of Jinkuishenqi pills had best effect. It is concluded that the mechanism of Jinkuishenqi pills on trea- ting mice with kidney- yang deficiency may be related to its protection effect on the testes and adrenal glands.
