聚乙二醇修饰对蛋白质类药物影响的研究进展 被引量:1

摘要 蛋白质类药物由于具有生物活性高、特异性好等优点,已成为一类重要的现代医药产品。然而在临床应用中,蛋白质类药物还存在一些问题,如半衰期较短,容易被循环系统清除;免疫原性较强,易引发变态反应;稳定性较差,容易被降解;水溶性较差,给药途径受限等。聚乙二醇(PEG)是一种无毒、高亲水性的大分子聚合物,PEG与蛋白质的非活性必需基团结合后,蛋白质药物的相对分子质量增加,表面的一些抗原决定簇被遮蔽,带电性质也会发生一定变化,这些改变通常可以使蛋白质类药物的稳定性增加、体内半衰期延长、免疫原性降低。
