冷冻液中添加硫代甘油对小鼠冷冻解冻精子体外受精的影响

Effect of adding monothioglycerol in cryoprotectant solution on in vitro fertilization of frozen-thawed mouse sperm

摘要 为了研究在冷冻液中添加硫代甘油(MTG)对小鼠冷冻精液解冻后受精能力的影响,试验采用在精液冷冻液中添加不同浓度MTG(160,320,480,640μmol/L)的方法冷冻C57BL/6小鼠和昆明白小鼠的精子,然后用解冻的精液进行体外受精,检测受精率和2-细胞发育率,还检测了添加480μmol/L组的C57BL/6小鼠体外受精胚胎移植后的出生率。结果表明:在冷冻液中添加MTG后C57BL/6小鼠和昆明白小鼠冷冻解冻精液的受精率和2-细胞发育率均显著升高(P<0.05),说明在冷冻液中添加MTG能够支持体外受精后正常的胚胎发育。通过添加MTG能显著提高这2个品系小鼠冷冻解冻后精子的体外受精能力。 To investigate the effect of adding monothioglycerol (MTG) in cryoprotectant solution on the fertilizing ability of frozen - thawed mouse sperm. The mouse sperm from Kunming and C57 BL/6 mouse strains was cryopreserved in the cryoprotectant solution with different con- centrations of MTG ( 160,320,480, and 640 μmol/L). The fertility rate and 2 - cell embryo development rate of the thawed sperm were deter- mined after in vitro fertilization using thawed semen, and the birth rate of C57 BL/6 mouse strain in the group adding 480 μmol/L MTG in cryo- protectant solution was also determined. The results showed that the fertility rates and 2 - cell embryo development rates of Kunming and C57 BL/ 6 mouse strains were significantly increased when adding MTG in cryoprotectant solution ( P 〈 0.05 ). MTG could support normal embryonic de- velopment after in vitro fertilization. The results indicate that adding MTG can significantly improve the fertilizing ability of frozen - thawed sperm from these two strains of mice.
