系统性红斑狼疮患者外周血单个核细胞CD28 mRNA的表达 被引量:3

Expression of CD28 mRNA in peripheral blood mononuclear cells of patients with systemic lupus erythematosus

摘要 目的 探讨CD2 8在系统性红斑狼疮 (SLE)患者外周血单个核细胞 (PBMC)中的表达水平及其意义。方法 应用反转录 聚合酶链反应 (RT PCR)检测了 34例活动期SLE患者和 30名正常人PBMC中CD2 8mRNA的表达水平。结果 活动期SLE患者CD2 8的阳性表达率为 2 0 6 % ,明显低于正常人对照组 (70 0 % ) ,差异非常显著 (P <0 0 0 1) ;活动期SLE组CD2 8的平均表达水平(0 19± 0 2 1)亦明显低于正常对照组 (0 43± 0 11) ,差异显著 (P <0 0 5 )。结论 CD2 8的异常表达可能在SLE发病机制中起作用 ,CD2 8mRNA的低水平表达可能与外周血CD2 8+ T细胞凋亡增加或迁移到炎症部位有关。 Objective To investigate the level of CD28 expression in patients with systemic lupus erythematosus (SLE) and its significance.Method Applying reverse transcription polymerase reaction (RT PCR) technique to semiquantitatively analyze CD28 mRNA expression in the peripheral blood mononuclear cells (PBMC) from 34 active SLE patients and 30 healthy subjects.Results The positive rate of CD28 expression in active SLE patients accounted for 20 6%,which was markedly lower than the normal controls (70 0%),the difference was statistically significant ( P <0 001).Compared with normal controls,the mean level of CD28 mRNA expression in active SLE group was decreased as well ( P <0 05).Conclusion The abnormal expression of CD28 may play a pathological role in the development of SLE.The lower expression of CD28 mRNA may contribute to the increased apoptosis of CD28 + T cells or the selective migration of these cells to sites of autoimmune injury in SLE patients.
