骨髓间充质干细胞修复兔椎间盘实验性研究

摘要 目的探讨骨髓间充质干细胞修复兔椎间盘损伤的效果。方法选取30只健康新西兰大白兔,随机分为观察组(n=15)和对照组(n=15)。2组兔均行椎间盘纤维环退行性变模型的建立。建模后观察组兔给予骨髓间充质干细胞与地塞米松磷酸钠的混合溶液进行治疗,对照组兔给予地塞米松磷酸钠进行治疗。对2组治疗后的椎间盘高度指数(DHI)、Ⅱ型胶原量及病理学情况进行比较。结果观察组兔在治疗后2、4、8、12周时的DHI和髓核组织的Ⅱ型胶原量与对照组比较差异均有统计学意义(P<0.05)。观察组兔病理分级优于对照组,差异有统计学意义(P<0.05)。结论骨髓间充质干细胞在体外经过培养能够有效地对椎间盘纤维环进行修复。 Objective To explore the effects of bone marrow mesenchymal stem cells for the repair of rabbit intervertebral disc. Methods Thirty healthy New Zealand white rabbits were randomly divided into observation group( n = 15) and control group( n = 15). The intervertebral disc degeneration model was established in the two groups. The observation group was given mixed solution of bone marrow mesenchymal stem cells and dexamethasone sodium phosphate treatment; the control group were given dexamethasone sodium phosphate treatment. The intervetebral disc height index( DHI),type Ⅱcollagen and pathology of the two groups were compared after treatment. Results There were statistic difference of DHI and type Ⅱcollagen at 2, 4,8,12 weeks after treatment between the observation group and control group( P 0. 05). Conclusion Bone marrow mesenchymal stem cells cultured in vitro can effectively repair intervertebral disc.
