温阳补肾方对去卵巢大鼠骨密度及骨标志物的影响

Effects of Wenyangbushen prescription on the bone mineral density and marker of bone in ovariectomized rats

目的探讨温阳补肾方对去卵巢大鼠骨密度及骨标志物的影响。方法取3月龄健康雌性SD大鼠40只,随机分为正常组、对照组、模型组和治疗组,每组10只。正常组大鼠行假手术,其余3组大鼠行双侧卵巢摘除术,术后30 d开始治疗,治疗组大鼠灌胃给予温阳补肾方,对照组大鼠给予戊酸雌二醇,治疗组及对照组大鼠各自按1 mL·100 g-1体质量灌胃;正常组和模型组大鼠每日按体质量给予同等剂量的生理盐水。于给药治疗12周后测定血清中骨原交联水平及Ⅰ型胶原N端前肽(PINP)、碱性磷酸酶(AKP)、钙和磷的水平;骨密度测量仪测骨密度。结果与正常组比较,模型组大鼠PINP水平明显升高,骨原交联水平下降,差异均有统计学意义(P<0.01);与模型组比较,治疗组大鼠骨原交联水平升高,PINP水平降低,差异均有统计学意义(P<0.01)。与对照组比较,模型组大鼠血清AKP水平降低,差异有统计学意义(P<0.05)。与模型组比较,治疗组大鼠血清AKP水平升高,差异有统计学意义(P<0.05)。与对照组和正常组比较,模型组大鼠骨密度明显下降(P<0.05);与模型组比较,治疗组大鼠骨密度明显升高,差异有统计学意义(P<0.05)。结论温阳补肾方能提高血清中AKP的水平,增加骨密度,抑制骨吸收,促进骨形成,具有防治骨质疏松症的作用。

Objective To study the effect of Wenyangbushen prescription on the bone mineral density（ BMD） and maker of bone in ovariectomized rats. Methods Forty three months old health female SD rats were selected and randomly divided into normal group,control group,model group and treatment group,with 10 rats in each group. The normal group was only shamly operated and the other three groups were ovariectomized. The rats were treated at 30 days after operation. The treatment group was given Wenyangbushen prescription（ 1 mL · 100 g- 1）; the control group was given estradiol valerate（ 1 mL·100 g- 1）; the normal group and model group were given isodose physiological saline according to the body weight per day. The levels of BMD,bone original crosslinking,procollagen type I N-terminal propeptide（ PINP）,alkaline phosphatase（ AKP）,calcium and phosphorus were determined at 12 weeks after treatment. Results Compared with normal group,the levels of PINP and bone original crosslinking in model group decreased（ P 0. 01）; compared with model group,the levels of bone original crosslinking increased and the levels of PINP decreased in treatment group（ P 0. 01）. The serum AKP in model group were significantly lower than those in control group and treatment group（ P 0. 05）. Compared with control group and normal group,the BMD in model group decreased（ P 0. 05）; compared with model group,the BMD in treatment group increased（ P 0. 05）. Conclusion The Wenyangebushen prescription can increase the level of AKP and BMD in ovariectomized rats; repress bone resorption and promote bone formation,thus it can be used to treat osteoporosis.
