几种组织培养上清液对造血祖细胞体外增殖分化的比较 被引量:2

摘要 目的:寻求既经济又有效的造血生长因子样活性物质。方法:采用造血祖细胞体外培养与造血生长因子生物活性检测等实验血液学技术,检测胎肌培养上清液、骨髓基质细胞培养上清液、胸腺细胞培养上清液与脾细胞培养上清液对造血祖细胞CFU-GM、BFU-E和CFU-Mix增殖分化的影响。结果:胎肌培养上清液和对CFU-GM和CFU-Mix的体外增殖与分化有明显刺激作用,而胸腺与脾细胞条件培养上清液对BFU-E的生长有显著促进作用。结论:胎肌培养上清液中可能含GM-CSF样活性物质,而胸腺与脾细胞培养上清液具有较高IL-3样活性,可以利用组织与细胞培养上清液替代重组造血生长因子进行造血细胞的实验研究和临床诊断。 Objective: To find some active materials that can be easily and cheaply attained and have similar function of hematopoietic growth factor(HGF). Methods: The technique of cell culture ex vivo and the assay of the biological activity of HGF were used. The supernatants of culture conditional media in fetal muscle tissue, thymocyte, spleen cell and human bone marrow stromal cell were collected. The effects of the different conditional media on the proliferation and differentiation of hematopoietic progenitor cell including CFU-GM, CFU-Mix and BFU-E were observed. Results: The culture conditional media of fetal muscle can markedly improve the proliferation and differentiation of CFU-GM and CFU-Mix; and the culture conditional media of thymocyte and spleen cell can enhance the growth of BFU-E. Conclusion: There may be some active materials similar GM-CSF in the supernatants of the bone marrow stromal cell and fetal muscle conditional media. The supernatants of thymocyte and spleen cell conditional media may function as the alike activity of IL-3. Such culture conditional media may act as the substitute for HGF in hematopoietic progenitor cell culture and in the clinic diagnostic assays.
