摘要 目的 采用HPLC-TOF/MS技术对青风藤不同提取部位及给药后大鼠血浆中的化学成分进行鉴别,以探索青风藤可能的药效成分。方法 色谱分离采用SHISEIDO MG C18(100 mm×3.0 mm,3μm)液相色谱柱,以乙腈-水(含0.1%的甲酸)为流动相,梯度洗脱,柱温20℃,流速0.3 mL.min-1。质谱定性采用飞行时间质谱,电喷雾离子源(ESI),正离子模式,质量数扫描范围m/z 100~1 000。结果 体外各部位共鉴别出青风藤中23个化学成分,大鼠血浆中共鉴别出6个入血成分。结论 该方法可用于青风藤体内外成分的鉴别,本研究为青风藤进一步的药效试验提供了参考和依据。 Objective To analyze the chemical components in different extracted parts of Caulis Sinomenii and in rat plasma after administration by HPLC-TOF/MS to investigate possible effective components of the herb. Methods The analyses were performed on a column ofSHISEIDO MG C18 (100 mm× 3.0 mm, 3 μm) with a mobile phase of acetonitrile and 0.1% formic acid at a gradient program. The temperature of the column was 20℃. The flow rate was 0.3 mL· min^-1. Time-of-flight mass spectrometer (TOF/MS) and electrospray ion source (ESI) were applied for qualitative analysis under positive ion mode, and mass scan range was m/z 100 - 1 000. Results Altogether 23 compounds in Caulis Sinomenii were identified, while 6 compounds were identified in the rat plasma. Conclusion The method is successfully used in identifying components in Caulis Sinomenii in vitro and in vivo, which provides a reference and evidence for further pharmacodynamics experiments.
