摘要
目的:建立母液中3-甲基黄嘌呤含量的测定方法。方法采用高效液相色谱法,分析柱为C18色谱柱,进行等度洗脱,流速1.0mL/min,检测波长:273 nm,柱温为35℃。结果3-甲基黄嘌呤浓度在100-800mg/L范围内呈良好线形关系,实验有较好的精密度,其RSD为0.21%。平均回收率为98.38%,RSD为0.29%。结论该检测方法操作性强、灵敏度高、准确、重复性好,可用于生产母液中3-甲基黄嘌呤的含量测定。
Objective To establish an HPLC method for determine of 3- methyl xanthine in mother liquor. Methods The chromatographic analysis was carried out using a Merck RP-18 colum at 35 ℃ , The mobile phase was 5g/L potassium dihydrogen phosphate solution- methanol (80 ∶ 20, V/V), at a flow rate of 1.0mL/min and UV detection at 273 nm. Results The line arrange was 100-800 mg/L, The precision is good with RSD of 0.21%. The average recovery of 3- methyl xanthine was 99.38% and RSD was 0.29%. Conclusion The method is simple, accurate and reproducible.
