Baicalein对人脐静脉血管内皮细胞12-脂氧化酶、血管内皮生长因子表达的影响

Effect of Baicalein on 12-LOX and VEGF Expression of Human Umbilical Vein Cells

摘要 目的:观察baicalein作用于人脐静脉血管内皮细胞系ECV-304细胞后12-脂氧化酶(12-LOX)、血管内皮生长因子(VEGF)表达的情况。方法:Baicalein处理培养的第2代血管内皮细胞,实验组分别用1,10,100μmol/L baicalein处理48h和30μmol/L的baicalein处理1,2,4,6d。设立空白对照组。RT-PCR检测各组12-LOX、VEGF的mRNA表达;Western blot免疫印记法检测各组12-LOX的表达水平。结果:RT-PCR扩增得到单一的产物,与预期的扩增序列大小完全一致,并且Western blot免疫印记法检测得出12-LOX的表达与baicalein的作用时间和作用浓度成反比。结论:血管内皮细胞自身能产生12-LOX、VEGF,它们均是促进新生血管的形成的因素,而baicalein抑制了12-LOX、VGEF的表达,从某一途径抑制新生血管的形成。 Objective:To investigate the effects of Baicalein on 12-1ipoxygenase (12-LOX) and vascular endothelial growth factor (VEGF) expression of human umbilical vein endothelial cell line ECV- 304. Methods. The second generation cultured vascular endothelial cells were exposed to com- plete medium as control, and medium containing 1, 10, 100 μmol/L Baicalein for 48 h, or medi- um containing 30 μmol/L Baicalein for 1, 2, 4, 6 d, respectively as experimental groups. RT PCR was performed to measure the mRNA expression levels related to VEGF and 12-LOX in different groups at different time points. The expression levels of 12-LOX were detected in different groups using the Western blot immunoblotting. Results: A single product was obtained by RT- PCR amplification, and its sequence and the size were consistent with expectations. For the 12- LOX expression levels assayed by the Western blot immunoblotting, the time of the exposed to Baicalein was inversely proportional to the concentrations of Baicalein. Conclusion:Vascular endo- thelial cells can produce 12-LOX and VEGF, which can enhance the neovascularizatiom Baicalein can inhibit the neovascularization by a specific pathway.
