摘要
目的观察滋阴活血方剂对糖尿病大鼠牙周组织的病理改变及VEGF表达的影响,探讨滋阴活血方剂对糖尿病牙周组织的作用。方法选用40只雄性Wistar大鼠,随机分为3组:正常组(10只)、糖尿病组(15只),滋阴活血方剂组(中药组)(15只)。糖尿病组、中药组腹腔注射STZ溶液,制备糖尿病大鼠模型。自建立糖尿病模型第一天起,中药组每只大鼠3ml/天(含生药1g/ml)中药灌喂;所有大鼠正常饮食水。持续喂养12周。对牙周组织切片进行HE染色和VEGF免疫组化染色,观察牙周组织的病理变化和VEGF表达的改变。结果糖尿病模型建立后,糖尿病组空腹血糖值[21.51±2.44mmol/L]和中药组空腹血糖值[21.76±1.82mmol/L]均高于正常组[5.51±0.75 mmol/L](P<0.05)。糖尿病组、中药组空腹血糖均大于15mmol/L,糖尿病模型建立成功。滋阴活血方剂干预12周后,中药组血糖[13.36±3.30mmol/L]下降(P<0.05),并低于糖尿病组[21.3±4.85mmol/L](P<0.05),均高于正常组[5.49±0.49 mmol/L](P<0.05)。中药组血糖水平[(13.36±3.30)mmol/L]显著低于糖尿病组[(21.3±4.85)mmol/L](P<0.05)。VEGF免疫组化平均光密度值(OD)糖尿病组[113.33±18.12]低于正常组[143.68±14.15]和中药组[132.39±10.66](P<0.05);正常组与中药组相比差异无统计学意义无差异(P>0.05)。结论滋阴活血方剂可降低大鼠实验性高血糖水平,并使其牙周组织中的VEGF表达水平趋于正常。
Objective To evaluate the effect of "Flow Nourishing Promoting Blood yin Recipe"(PBFNR) on the pathological changes and the expression of Periodontium VEGF in rat with diabetes. Methods Forty male Wistar rats were randomly divided into three groups including normal control group (group NG, rl=10), diabetes group (group DM, n=15) and PBFNR group (group NR, n=15). The rat diabetic model was produced by injection of streptozotoein (STZ,45mg/kg) in group DM and group NR. PBFNR was administered at a dose of 1g/ml in group NR for 12 weeks, 3ml per day. The maxilla was then dissected under anesthesia. The tissue sections were stained with HE for histological examination,and the VEGF expression was detected by immunohistochemical method. Results The average fasting blood glucose in group NG [5.51±0.75mmol/L] was lower than that in group DM [21.51±2.44 mmol/L] and group NR [21.76± 1.82 mmol/L] (P〈0.05). The model rat blood glucose were exceed 15mmol/L, animal model of diabetes established successfully. At 12 weeks post-treatment, the blood glucose levels in group NR [13.36±3.30mmol/L] decreased, which was lower than that in group DM [21.3±4.85mmol/L] (P〈0.05). Both in group DM and NR ,the blood glucose levels were higher than that of group NG [5.49±0.49 mmol/L](P〈0.05 ). The average optical density value (OD) of the expression of VEGF in group DM [113.33±18.12] was lower than that in group NG [143.68±14.15] and group NR [132.39±10.66] (P〈 0.05), there was no significant difference between group NG and group NR (P〉0.05). Conclusion PBFNR could decrease high blood glucose levels in experimental diabetes rats and decrease VEGF expression to a nomal level in periodontium.
