大鼠肠系膜淋巴结在生后发育过程中高内皮微静脉的结构和形态

Structure and morphology of the high endothelial venules of the rat mesenteric lymph node during the postnatal development

摘要 目的研究生后大鼠肠系膜淋巴结内高内皮微静脉(high endothelial venules,HEV)的发育过程,为探讨其发生发育机制提供形态学依据。方法应用透射电镜观察出生后第0天、2天、6天、7天、8天、10天、12天、14天、18天、21天、25天、28天、31天、35天、40天的新生大鼠肠系膜淋巴结内高内皮微静脉的结构和形态;应用免疫组织化学和免疫荧光染色方法观察各日龄大鼠肠系膜淋巴结组织中高内皮微静脉的分布。结果从第0天到第6天大鼠肠系膜淋巴结内无典型的高内皮微静脉,内皮核不明显高大,属于类高内皮微静脉,肠系膜淋巴结外周区可见少量类高内皮微静脉,中心区则未见类高内皮微静脉。第7天肠系膜淋巴结外周区高内皮微静脉数量增多,但其分布仍局限于周边区,到第10天才出现典型的高内皮微静脉,内皮核高大呈立方形。结论生后大鼠肠系膜淋巴结内高内皮微静脉的发育规律为探讨其发生发育机制提供了形态学依据。 Objective To study the structure and nlorphology of high endothelial venules of the rat rnesenteric lymph nodes during postnatal development to provide the morphological evidence for the oecmvence and development mechanism of the high endothelial venules. Methods The mesenteric lymph nodes from the postnatal different-age rats (0d, 2d, 6d, 7d, 8d, 10d, 12d, 14d, 18d, 21d, 25d, 28d, 31d, 35d, 40d) were embedded and sectioned. The structure and morphology of the high endothelial venules were observed by transmission electron microscope. Irnmunohistochemistry and immunofluorescenee were used to observe the distribution of the high endothelial venules in mesenteric lymph nodes. Results In animals from 0 day to 6 days old, no high endothelial venules could be found, the high endothelial venule-like blood vessels were distributed in the peripheral of the mesenteric lymph nodes, not in central part of the node. The nnmber of high endothelial venules in peripheral area increased at 7 days after birth, the typical high endothelial venules were found with euboidal nuclear of high endothelial venules at 10 days after birth. Conclusion The present results would provide the morphological evidence for the further study of occurrence and development mechanism of the high endothelial venules.
