大鼠白介素-1β在异氟烷预处理脊髓损伤中的作用 被引量:1

The effect of interleukin-1β on spinal cord injury after isoflurane preconditioning in rats

摘要 目的探讨白介素-1β(IL-1β)在异氟烷预处理大鼠脊髓损伤(spinal cord injury,SCI)时的作用。方法:雄性成年SD大鼠75只,体重250—300 g,将大鼠随机分为3组(n=25):假手术组(S组)、脊髓损伤组(SCI组)、异氟烷预处理组(ISO组)。采用打击法制作脊髓损伤模型。ISO组吸入2%异氟烷2 h,24 h后制备脊髓损伤模型。连续7 d采用BBB评分进行神经功能缺陷评分,免疫荧光标记神经元特异性核蛋白(neuron-specific nuclear protein,NeuN)和白介素-1β的表达,其余动物处死,采用Western Blot法测定白介素-1β的表达水平。结果:与S组比较,SCI组和ISO组神经功能缺陷评分降低,NeuN和IL-1β表达增加,IL-1β蛋白表达上调(P〈0.05);与SCI组比较,ISO组神经功能缺陷评分增高,NeuN和IL-1β表达下降,IL-1β蛋白表达下调(P〈0.05)。结论:异氟烷预处理可能通过抑制IL-1β表达,从而减轻大鼠脊髓损伤。 Objective: To explore the effect of interleukin-lbeta( IL-1β) on spinal cord injury( spinal cord injury, SCI) after isoflurane preconditioning in rats. Methods : Seventy-five adult male SD rats, weighing 250 - 300 g were randomly divided into three groups (n =25) : sham operation group (S group), spinal cord injury group (SCI group), isoflurane preconditioning group (ISO group). Spinal cord injury model was made by percussion. Inhalation of 2% isoflurane for 2 h in rats of ISO group was performed to make SCI 24 h post treatment. Neurological deficit scores were evaluated by BBB score for consecutive seven days, the expression of NeuN (neuron-specific nuclear protein) and IL-1β were labeled by immunofluorescence, the rest rats were killed and the expression of IL-1β protein was detected by Western Blot. Results: Compared with S group, the neurological deficit score was significantly lower. The expression of NeuN and IL-1β was increased and IL-1β protein was up-regulated in SCI and ISO group( P 〈 0.05 ). Compared with SCI group,the neurological deficit score was significantly enhanced, the expression of NueN and IL-1β was reduced, IL-1β protein was down-regulated in ISO group ( P 〈 0.05 ). Conclusion : Isoflurane preconditioning could reduce spinal cord injury by inhibition of IL- 1β.
