摘要
目的:观察4周离心运动及针刺干预对生肌调节因子(MRFs)的影响,探讨MRFs变化特征及针刺在骨骼肌损伤修复中的作用。方法:72只雄性SD大鼠分为安静组(C)、运动组(E)和运动针刺组(EA),E与EA组进行坡度为-16°的4周跑台运动;免疫印迹法检测生肌调节因子MyoD、Myf5、Myogenin、MRF4蛋白表达。结果:第1、3、4周E组MyoD、MRF4蛋白表达显著低于C组和EA组(P<0.05);EA组MyoD蛋白表达第1、3、4周接近安静水平,MRF4蛋白表达第4周显著低于C组(P<0.05)。E组Myf5蛋白表达呈上升趋势,第4周显著高于C组(P<0.05);EA组Myf5蛋白表达第1至3周均高于E与C组;E组Myogenin蛋白表达持续升高,且第3、4周显著高于C组(P<0.05);EA组Myogenin蛋白表达第1、2周显著高于E组(P<0.05),第3、4周与之相反。结论:4周离心运动能上调Myf5与Myogenin蛋白表达,下调MyoD与MRF4蛋白表达;针刺具有调控骨骼肌MRFs保持正常水平的作用。
Objective The aim of this study is to observe the effects of four-week eccentric exercise and acupuncture on the myogenic regulatory factors(MRFs). Methods 72 male SD rats were divided into three groups:control group (C),exercise group (E)and exercise plus acupuncture group (EA). Rats in groups E and EA were submitted to four-week downhill running (-16°)on a treadmill for 4 weeks. Rats in groups EA received acupuncture therapy immediately after running. Protein expression of MyoD, MyfS,myogenin and MRF4 were detected by western blot. Results The MyoD and MRF4 protein in group E decreased significantly comparing to groups C and EA in the lst,3rd,and 4th week(P 〈 0.05). MyoD protein in group EA was close to normal level in the lst,3rd,and 4th week,and MRF4 protein significantly lower than control group in the 4th week (P 〈 0.05). Myf5 protein in group EA was higher than groups E and C from the 1st to 3rd week. Myogenin protein in group E went up steadily and was significantly higher than in group C in the 3rd and 4th week (P 〈 0.05). Myogenin protein in group EA was higher than in group E in the 1st and 2nd week(P 〈 0.05) ,but was inversely lower than in group E in the 3rd and 4th week (P 〈 0.05). Conclusion Four-week eccentric exercise may up-regulate Myf5 and myogenin protein expression,and down-regulate MyoD and MRF4 protein expression,meanwhile acupuncture can maintain normal levels of MRFs.
