鸡血藤中总黄酮的聚酰胺树脂纯化工艺优选 被引量:6

Optimization of Purification Technology of Total Flavonoids from Spatholobi Caulis by Polyamide Column Chromatography

摘要 目的:优选鸡血藤总黄酮的聚酰胺树脂纯化工艺。方法:采用UV测定总黄酮含量,检测波长278nm。选择聚酰胺柱层析法,以总黄酮含量和转移率为指标,通过单因素试验考察上样液质量浓度、吸附流速、洗脱剂用量等对鸡血藤总黄酮纯化工艺的影响。结果:最佳纯化工艺为上样量9.076mg·g-1,上样液质量浓度1.261g·L-1,吸附、洗脱速度2BV·h-1,加水5BV和20%乙醇3BV洗脱除杂,加70%乙醇4BV洗脱,收集第2~4BV洗脱液;总黄酮纯度达81.5%,转移率77.5%。结论:该纯化工艺稳定可行且重复性好,可有效富集鸡血藤总黄酮部位。 Objective: To optimize purification technology of total flavonoids from Spatholobi Caulis by polyamide column chromatography. Method: UV was adopted to determine the content of total flavonoids with detection wavelength at 278 nm. Polyamide column chromatography was used, taking content and transfer rate of total flavonoids as indexes, single factor tests were adopted to optimize purification process with the concentration of sample solution, velocity of adsorption, eluent dosage and so on as factors. Result: Optimum purification technology was as following: sample volume of 9. 076 mg ·g-1 the concentration of sample solution 1. 261 g ·L-1 adsorption and elution velocity of 2 BV ·h-1, eluted impurity with 5 BV of water and 3 BV of 20% ethanol, then eluted with 4 BV of 70% ethanol, collected eluent of the second-fourth BV; purity and transfer rate of total fiavonoids were 81.5% and 77.5% , respectively. Conclusion: This purification technology was stable and feasible with good repeatability, it could enrich portion of total flavonoids effectively.
