健脾益肠散对溃疡性结肠炎大鼠MyD88的影响

Effect of Jianpi Yichang Powder on MyD88 in Ulcerative Colitis Rats

目的:探讨健脾益肠散对溃疡性结肠炎(UC)大鼠髓样分化因子88(MyD88)的影响。方法:将60只大鼠随机分为正常组、模型组、柳氮磺吡啶组(0.3 g·kg-1)及健脾益肠散高(204 g·kg-1)、中(136 g·kg-1)、低剂量(68 g·kg-1)组,每组10只;除正常组外,其余均采用2,4,6-三硝基苯磺酸(TNBS)/乙醇法复制UC大鼠模型;灌胃给药21 d后,用ELISA、免疫组化、Western blot及RT-PCR法检测各组大鼠血清MyD88含量和结肠组织MyD88的表达。结果:模型组大鼠血清MyD88水平及结肠组织MyD88蛋白和mRNA表达均明显高于正常组(P<0.01);健脾益肠散高、中剂量组血清MyD88含量和结肠MyD88蛋白及mRNA表达均较模型组降低,差异有统计学意义(P<0.05或P<0.01);健脾益肠散低剂量组MyD88蛋白表达亦低于模型组(P<0.05);健脾益肠散高剂量组对MyD88的影响与柳氮磺吡啶组比较差异不显著,但明显优于低剂量组(P<0.05)。结论:健脾益肠散对UC肠黏膜免疫的保护作用可能与降低外周血MyD88水平及结肠组织MyD88的蛋白和基因表达有关。

Objective： To explore the effect of Jianpi Yichang powder （JP） on myeloid differentiation factor 88 （ MyD88 ） in ulcerative colitis （ UC ） rats. Method ： Sixty rats were randomly divided into the normal group, model group, sulfasalazine group （0.3 g ·kg-1 ）, JP high （204 g ·kg-1 ）, medium （ 136 g ·kg-1 ） and low dose （68 g·kg-1 ） group of 10 each. Besides normal group, the other were used 2, 4, 6-trinitrobenzene sulfonic acid （TNBS） /ethanol method copied UC rat model. Observed the serum of MyD88 level and colon tissue MyD88 expression were respectively detected by ELISA, immunohistochemistry, Western blot and RT-PCR in each group of rats after intragastric administration 21 d. Result： The model rats serum of MyD88 level and colon tissue MyD88 protein and mRNA expression significantly higher than the normal group （P 〈 0. 01 ）. Serum of MyD88 level and colon tissue MyD88 protein and mRNA expression in JP high, middle dose group compared with modelgroup decreased significantly （ P 〈 0. 05 or P 〈 0. 01 ）. MyD88 protein expression in JP low dose group was also lower than model group （P 〈 0. 05）. JP high-dose group, the difference of MyD88 with sulfasalazine group was not significant, but significantly better than the low-dose group （P 〈 0. 05 ）. Conclusion： Protective effects of JP on intestinal mucosal immunity of UC may be related to the decrease of MyD88 level and its protein and gene expression.
