摘要
目的:D-半乳糖sc建立亚急性衰老小鼠模型,研究豆丹的抗衰老作用。方法:将实验动物随机分为7组,分别为正常对照组、衰老模型组、西洋参组(0.5 g·kg-1)、豆丹水提液高(2.8 g·kg-1)、低剂量组(1.4 g·kg-1)、豆丹醇提液高(12.5 g·kg-1)、低剂量组(6.25 g·kg-1)。造模14 d后开始给药,给药14 d后测定负重游泳时间、脏器系数、肝脏超氧化物歧化酶(SOD)活性、肝脏及脑组织谷胱甘肽过氧化物酶(GSH-Px)活性、肝及脑组织丙二醛(MDA)含量。结果:与正常组比较,衰老模型组负重游泳时间、脏器指数、SOD及GSH-Px均降低,MDA含量增加,且均有显著性差异(P<0.05或P<0.01);与衰老模型组比较,豆丹水提液、醇提液均能显著延长负重游泳时间(P<0.05或P<0.01),提高脏器指数(P<0.05),提高SOD,GSHPx活性(P<0.05或P<0.01),降低MDA含量(P<0.05),且对雌雄小鼠的作用大体相当。结论:豆丹可能通过提高机体免疫、抗脂质过氧化和清除氧自由基的能力来发挥抗衰老的作用。
Objective: Anti-aging effect of Clanis bilineata tsingtauica was investigated through the model of subacute aging mice induced by subcutaneous injection of D-galactose in the study. Method: The Kunming mice were divided into seven groups: normal control group, aging model group, Panax quinquefolius positive control group, low and high-dose C. bilineata tsingtauica's water extraction group (1.4, 2.8 g ·kg-1) , low and high-dose Clanis bilineata tsingtauica's 75% ethanol extraction (6.25, 12.5 g ·kg-1 ). 14 days after the mice were sc given D-galactose, the corresponding drugs were ig given to the mice, respectively. After 14 days, the loaded swimming time, the viscera index, superoxide dismutase (SOD) activities in liver homogenate, glutathione peroxidase (GSH-Px) activities in liver and brain homogenate and malondialdehyde (MDA) content in liver and brain homogenate were measured. Result: Compared with the normal control group, the loaded swimming time, the viscera index, the T-SOl) activities and the GSH-Px activities in liver and brain were markedly reduced in aging model group, while the content of MDA was markedly increased (P 〈 0.05 or P 〈 0.01 ). Both water extraction and 75 % ethanl extraction of C. bilineata tsingtauica could significantly extend the loaded swimming time (P 〈 0. 05. or P 〈 O. 01 ), increase the viscera index (P 〈 0. 05), elevat the activities of T-SOD and GSH-Px and decrease the contents of MDA ( P 〈 0.05 or P 〈 0.01 ) , and the effects on male and female are generally the same. Conclusion: C. bilineata tsingtauica has an anti-aging effect by improving immune and antioxidant capacity.
