摘要
目的:观察葶苈草颗粒对炎症性肠病(IBD)的治疗作用并探讨其作用机制。方法:选昆明种小鼠,用3%恶唑酮(OXZ)复制小鼠OXZ结肠炎模型。实验分为正常对照、模型对照组(等容蒸馏水)、葶苈草颗粒(1.95,3.9,7.8 g·kg-1)治疗组、柳氮磺胺吡啶(0.39 g·kg-1)阳性药对照组,ig每天1次,连续治疗4 d。用酶联免疫吸附(ELISA)法测定血清和结肠组织中白细胞介素-10(IL-10)、结肠组织中髓过氧化物酶(MPO)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)的含量,进行疾病活动指数(DAI)评分和病理组织学观察。结果:治疗4 d后,与模型组比较,葶苈草颗粒3.9,7.8 g·kg-1治疗组,血清和结肠组织IL-10含量明显升高(P<0.05或P<0.01),结肠组织MPO,IL-1β,TNF-α和IL-6含量明显降低(P<0.05或P<0.01);葶苈草颗粒治疗组总DAI值明显减少(P<0.05或P<0.01)。结论:葶苈草颗粒对OXZ诱导的小鼠实验性肠炎具有较好的治疗作用,其可能通过调节促炎细胞因子IL-1β,TNF-α,IL-6和抑炎细胞因子IL-10之间的平衡,有效减轻实验性结肠炎炎症,从而起到治疗IBD的作用。
Objective: To explore the therapeutic effect of pepperweed seed on inflammatory bowel disease (IBD) and the underlying mechanism. Method: The IBD mice model was induced by oxazolone (OXZ). Fifty modeling mice were randomly divided into 5 groups : model group, sulfasalazine ( SASP, 0.39 g · kg- 1 ) group and pepperweed seed (1.95, 3.9, 7.8 g·kg-1) groups, 10 mice each group. Another 10 mice were selected as normal group. The modeling mice were orally treated daily for 4 days. The normal group and the model group received distilled water for 4 days. The contents of interleukin (IL) -10 in serum and colon were detected by enzyme-linked immunosorbent assay (ELISA). Similarly, the contents of myeloperoxidase (MPO) , interleukin IL-1β, IL-6 and tumor necrosis factor-α (TNF-a) in colon tissues were detected. The disease activity index (DAI) and histological changes of colon were evaluated. Result: After 4 days' treatment, compared with the model group, the contents of interleukin 10 (IL-10) in serum and colon tissues in pepperweed seed (3.9, 7.8 g · kg- 1 ) groups were significantly increased ( P 〈 O. 05, P 〈 O. O1 ), the contents of MPO, 1β( IL-1β), IL-6 andTNF-α in colon tissues in pepperweed seed (3.9, 7.8 g ·kg-1) groups were significantly reduced. The total DAI level in pepperweed seed groups were significantly decreased (P 〈 0.05, P 〈 0.01 ). Conclusion: The pepperweed seed exerted therapeutic effect on oxazolone-induced colitis in mice, which is related to the regulation of IL-1β, TNF-α, IL-6 and IL-10. It can alleviate oxazolone-induced colitis and be valuable in the IBD treatment.
