摘要 为研究AS1411对宫颈癌HeLa细胞的辐射增敏机理,采用γ-H2AX免疫荧光法观察HeLa细胞DNA损伤情况;流式细胞术检测AS1411对HeLa细胞周期及凋亡的影响。结果显示:经AS1411预处理的HeLa细胞核内DNA断裂点数目明显增多;AS1411预处理不影响由辐射引起的细胞周期阻滞,但能够促进辐射诱导的HeLa细胞凋亡率的增长。AS1411通过阻止由辐射引起的DNA损伤的修复及诱发细胞凋亡增加HeLa细胞的辐射敏感性,但此作用机理与细胞周期无关。 The aim is to investigate the effects of AS1411 on radiosensitivity and the relative mechanism on uterine cervical cancer HeLa cells, γ-H2AX immunofluorescence technique was applied to observe the DNA damage of HeLa cells; change in cell cycle and cell apoptosis rate was detected by flow cytometry. The results show that γ-H2AX loci of HeLa cell increase significantly after being pretreated by AS1411; AS1411 pretreatment does not influence the cell cycle arrest caused by radiation, but it does accelerate the apoptosis rate of HeLa cells induced by radiation. By increasing the DNA double-strand breaks and inducing cell apoptosis of the HeLa cells caused by radiation, AS 1411 can enhance the radio sensitization of HeLa cells. However, the mechanism of the radiosensitizing effect is uncorrelated with cell cycle.
