摘要 考察氧载体对恩拉霉素发酵合成的影响,筛选出适合恩拉霉素发酵的氧载体并优化其添加方式。首先以恩拉霉素含量和菌体浓度作为评价指标,考察不同氧载体对恩拉霉素发酵的影响,确定最合适的氧载体。之后采用单因素试验法考察最适氧载体添加时间和添加量,最后通过响应面法优化最适氧载体的添加方式。结果表明:正十六烷为最适氧载体,其最优添加量为0.72%,添加时间为44h。该条件下进行发酵可使生产菌菌体浓度较对照提高23%,恩拉霉素含量达到2 940.65mg/L,与理论预测值的相对误差在0.48%,较对照提高70%。因此,添加正十六烷能加强发酵液中氧传递,提高溶氧水平,从而促进菌体生长及恩拉霉素合成。 The influences of oxygen vector on fermentation production of enramycin was studied. Meanwhile, the most effective type of oxygen vectors was identified and its adding concentration and timing were optimized. First, cell volume and the content of enramycin were used to identify the effectiveness of oxygen vectors. Then different concentrations of the optimum oxygen vector were added into the medium at different times during a single factor experiment. The fitting potency response surfaces were decided statistically using experimental data. The results showed that n-hexadecane was the most suitable oxygen vector. The optimum concentration of n-hexadecane was 0.72% and the best adding time was 44 h. Under the optimum conditions, the cell volume was increased by 23%, the content of enramycin was up to 2 940.65 mg/L, with the theoretical prediction relative error in 0. 48%, 70% higher thancontrol. In conclusion, n-hexadecane is an effective oxygen vector to enhance oxygen delivery, which can increase cell volume and enramvcin oroduetion.
