摘要
试验旨在研究流加培养基中不同营养成分、初始接种密度、流加起始时间对BHK21悬浮细胞流加培养的影响。结果表明,流加浓缩包中的氨基酸对促进细胞生长有明显影响,而无血清培养基添加成分不仅促进细胞生长,且对细胞活力的维持起着较为重要的作用。初始接种密度为3×10^5~6×10^5/mL,培养至48h开始流加一定量的浓缩液,流加培养效果较好。另外,在已优化的条件下进行流加培养,未出现细胞大量凋亡现象。
The research was aimed to study the impacts of different feeding nutrients, start time of feeding and cell inoculation density on the growth of BHK-21 suspension cells. The result showed that amino acid concentrated liquid had a apparent influence on promoting the growth of BHK-21 suspension cells. But serum components played a more important role in the promotions of cell growth, maintenance and cell activity. When initial inoculation density was 3 × 10^5 to 6× 10^6 cells/mL, adding a certain amount of concentrated liquid at 48 h, BHK-21 suspension cells grew well. In addition, BHK-21 suspension cells did not obviously occur apoptosis in the optimized conditions.
