过量氟对大鼠血清过氧化物抗氧化物及骨和软骨RNA含量的影响 被引量:7

摘要 目的 探讨脂质过氧化物、抗氧化物质水平和骨、软骨 RNA合成的变化在氟骨症发生机制中的作用。方法 将 6 5只 4~ 5周龄 ,体重 5 0~ 90 g的 SD大鼠随机分为 4组。用化学比色法测定血清超氧化物歧化酶(SOD) ,硫代巴比妥酸法测定血清丙二醛 (MDA) ,酶动力法测定血清碱性磷酸酶 (AL P) ,异硫氰酸胍法测定骨和软骨 RNA。结果  1血清 SOD在染氟后 3个月显著下降 ,5个月时明显恢复 ;2染氟后 5个月高氟组血清 MDA明显升高 ,有随尿氟排泄量增高而增高的趋势 ;3高氟组血清 AL P在实验后 3个月明显升高 ;4摄氟组大鼠骨、软骨 RNA含量均较对照组低 ,有随染氟剂量增加而减少的趋势。结论 过量氟实验大鼠早期体内自由基增多 ,抗氧化能力下降 ,骨、软骨 RN A含量减少 ,提示氟骨症早期摄入过量氟可直接影响骨与软骨 Objective Role of lipid peroxidation (lpo),antioxidation and RNA content in bone and cartilage on the pathogenesis of the skeletal fluorosis has been investigated in this paper.Methods 65 SD rats,4~5 week old,body weight 50~90g,were divided randomly into four groups.SOD,MDA and ALP in serum were determined by chemical colorimetry,sulfobabitoric acid and IFCC recommend methods,respectively,and RNA in bone and cartilage by guanidine thiocyanate method.Results ①Serum SOD in rats treated with excessive fluoride was decreased in three months after the starting of the experiment and recovered obviously in five months after the experiment.②Serum MDA in high fluoride group was increased obviously in three months after the starting of the experiment,and increase when exposed to high level of fluoride for five months continuoully.There was an increased trend of MDA in serum with the increased urine fluoride excreted.③Serum ALP in rats of groups treated with excessive fluoride was increased in three months after the starting of the experiment.④RNA content in bone and cartilage of rats treated with excessive fluoride was lower than the control,and a decreased trend of RNA content in bone and cartilage with a increased dose of fluoride was found.Conclusions SOD activities in serum and RNA content in bone and cartillge were decreased,and MDA and ALP activities in serum increased in rats treated with excessive fluoride.It meant that the free radical was increased and the antioxidation decreased in early stage of skeletal fluorosis.The decrease of RNA content in bone and cartillage showed excessive fluoride influenced RNA synthesis of bone and cartilage directly in the early stage of skeletal fluorosis.
