灰兜巴多糖的提取分离纯化研究 被引量:1

Extraction, isolation and purification of Huidouba polysaccharides

摘要 目的:对灰兜巴多糖进行提取、分离及纯化研究,为其做进一步的结构分析和对糖尿病的治疗提供新的线索。方法:采用水提醇沉法提取灰兜巴多糖;同时用Sevage法、木瓜蛋白酶-Sevage法和三氯乙酸法对其进行脱蛋白效果比较;最后用DEAE-52纤维素离子交换层析分离得到组分。结果:分别用苯酚-硫酸法和考马斯亮蓝G-250法测得灰兜巴粗多糖中的多糖含量为28.02%和蛋白质含量为1.23%;木瓜蛋白酶-Sevage法的蛋白质脱除率最高为96.08%;通过DEAE-52纤维素离子交换层析得到4个组分,总的多糖回收率为86.22%。结论:综合考虑选择木瓜蛋白酶-Sevage法为灰兜巴粗多糖脱蛋白的最佳工艺,HDB-2的多糖回收率较其他3个组分的要高,表明其溶解性较好。 Objective: Study on the extraction, isolation and purification of Huidouba polysaccharides, to provide new clues for further structural analysis and treatment of diabetes. Method: We used the water extraction and alcohol precipitation method to extract polysaccharides from Huidouba; Compared the effect of Sevage method, Papain-Sevage method and Trichloroacetic acid method on deproteinization. Huidouba polysaccharides were isolated by DEAE-Cellulose 52. Result: Respectively used phenol-sulfuric acid method and Coomassie brilliant blue G-250, The content of polysaccharide and protein is 28.02% and 1.23% in Huidouba polysaccharides. Protein removal rate of Papain-Sevage method up to 96.08%. We obtained four components through DEAE-Cellulose 52, the total polysaccharides recovery of 86.22%. Conclusion: Papain-Sevage method is the best method for Huidouba polysaccharides on deproteinization, polysaccharide recovery of HDB-2 is more high than other three components.
