以牙鲆(Paralichthys olivaceus)为母本,圆斑星鲽(Verasper variegatus)为父本进行科间远缘杂交,利用ISSR标记对2个家系杂交子代进行扩增,分析了双亲位点在杂交子代中的传递分离方式以及双亲与杂交子代的遗传距离。结果表明:6个引物在0401#和0402#2个家系中扩增的位点数为136和144个,其中多态性位点数量为90和91个,分别占66.18%和63.19%,2个家系间没有明显差异。2个家系中双亲与杂交子代的共有位点数量分别达34.56%和38.89%,表明遗传物质传递给了杂交子代,证明了科间远缘杂交的成功。2个家系中符合孟德尔分离规律的位点分别占32.35%和22.92%,出现分离但偏孟德尔分离位点占23.53%和27.78%,2个家系中均观察到部分非亲位点。在这2个家系中,杂交子代与母本牙鲆的遗传距离分剐为0.2941、0.3182,而与父本星鲽的遗传距离分剐为0.3645、0.3750,表明杂交子代在遗传上偏向母本牙鲆。该研究结果为进一步利用杂交群体同时构建牙鲆和星鲽的遗传连锁图谱提供了重要参考。
Distant hybridization between Paralichthys olivaceus females and Verasper variegatus maleswere conducted. ISSR markers were used to amplify the genomic DNA of both the parents and F1 hybrids. The polymorphic status of these markers in the progeny, the inheritance and segregation patterns from parents to their hybrids, and genetic distance between parents and F1 were analyzed. Sixprimers amplified 136 and 144 loci from families 0401# and 0402# , respectively, of which 90 (66.18%)and 91 (63.19%) were polymorphic. There was no significant difference between the two families. Thepercentages of loci inherited from parents to F1 hybrids were 34. 56% and 38.89%. These loci provedthe successful transmission genetic materials from both parents to the hybrids, indicating the success ofhybridization. A sum of 32.35% and 22.92% of loci in the progeny of the two families segregated in accordance with the law of Mendelian inheritance. In comparison, 23.53% and 27.78% of loci showeddeviation from Mendelian inheritance mode. In addition, some proportions of non-parent loci were observed in both families. The genetic distances between the mother and the progeny were 0. 2941 and 0. 3182, whereas between fathers and the progenies were 0. 3645 and 0. 3750 in the two families, respectively, indicating that the F1 hybrids were genetically closer to mother P. olivaceus. The above results can provide certain basis for the simultaneous construction of linkage maps of P. olivaceus and V.variegate using the hybrid as mapping population.
